- Abstract CYP2C19 is a member of the human microsomal cytochrome P450 (CYP). Significant variation in CYP2C19 levels and activity can be attributed to polymorphisms in this gene. Wildtype CYP2C19 and 13 mutants (CYP2C19.1B, CYP2C19.5A, CYP2C19.5B, CYP2C19.6, CYP2C19.8, CYP2C19.9, CYP2C19.10, CYP2C19.11, CYP2C19.13, CYP2C19.16, CYP2C19.19, CYP2C19.23, CYP2C19.30, and CYP2C19.33) were coexpressed with NADPH‐cytochrome P450 reductase in Escherichia coli. Hydroxylase activity toward testosterone and progesterone was also examined. Ten CYP2C19 variants showed Soret peaks (450 nm) typical of P450 in the reduced CO‐difference spectra. CYP2C19.11 and CYP2C19.23 showed higher testosterone 11α, 16α‐/17‐ and progesterone 6β‐,21‐,16α‐/17α‐hydroxylase activities than CYP2C19.1B. CYP2C19.6, CYP2C19.16, CYP2C19.19, and CYP2C19.30 showed lower activity than CYP2C19.1B. CYP2C19.9, CYP2C19.10. CYP2C19.13, and CYP2C19.33 showed different hydroxylation activities than CYP2C19.1B. - The strawberry fruit contains abundant polyphenols, such as anthocyanins, flavan-3-ol, and ellagitannin. Polyphenol enrichment improves the quality of strawberries and leads to a better understanding of the polyphenol induction process. We measured the total polyphenol content of strawberry fruits under different growth conditions, developmental stages, and treatment conditions during pre-harvest and post-harvest periods. High fruit polyphenol content was observed in cold treatment, which was selected for further analysis and optimization. A transcriptome analysis of cold-treated fruits suggested that the candidate components of polyphenols may exist in the phenylpropanoid pathway. Coverage with a porous film bag excluded the effects of drought stress and produced polyphenol-rich strawberry fruits without affecting quality or quantity. The degree of stress was assessed using known stress indicators. A rapid accumulation of abscisic acid was followed by an increase in superoxide dismutase and DPPH (2,2-Diphenyl-1-picrylhydrazyl) activity, suggesting that the strawberry fruits responded to cold stress immediately, reaching the climax at around 6 days, a trend consistent with that of polyphenol content. - African violet (Saintpaulia ionantha) is an herbaceous perennial of the Gesneriaceae family. Because almost all the cultivars are heterozygous, pure lines are useful for both classical and new breeding approaches. - To improve several agronomic traits in crisphead lettuce (Lactuca sativa L.) under high-temperature growth conditions, we investigated the correlation among those traits in multiple cultivars and performed genetic mapping of their causal genes. In a field cultivation test of Empire type (serrated leaf) and Salinas type (wavy leaf) cultivars, Empire type cultivars showed increased tipburn susceptibility and late bolting compared with Salinas type cultivars. We attempted genetic mapping of leaf shape and bolting time by ddRAD-seq using the F2 population derived from a cross between 'VI185' (Empire type) and 'ShinanoGreen' (Salinas type). These analyses suggested that both traits are controlled by a single locus in LG5. Genotyping of 51 commercial lettuce cultivars with a tightly linked marker (LG5_v8_252.743Mbp) at this locus showed an association between its genotype and the serrated leaf phenotype. By further fine mapping and transcriptome analysis, a gene encoding putative CIN-like TCP transcription factor was determined to be a candidate gene at this locus and was designated as LsTCP4. An insertion of retrotransposable element was found in the allele of 'VI185', and its transcript level in the leaves was lower than that in 'ShinanoGreen'. Because shapes of leaf epidermal cells in 'VI185' were similar to those in the TCP family mutant of Arabidopsis thaliana, the leaf shape phenotype was likely caused by reduced expression of LsTCP4. - The purpose of this study was to develop a hyperspectral imaging system that can detect gene recombinant calluses. Drug tolerance genes are commonly used as marker genes, although they may negatively affect the environment. Hence, we aimed to use the genes that code for nitrate reductase (NR) as marker genes. Once the NR genes are introduced into a cell, the recombined genes would show greater nitrate reduction compared with the wild-type genes, making it possible to use the nitrate concentration for discriminating gene recombinant calluses from non-recombinant ones. In the present study, a non-destructive method of measuring the nitrate concentration in spinach (Spinacia oleracea L. 'Orai') calluses was developed to allow for the rapid detection of the gene recombinant calluses. Absorption spectra of 349 calluses (concentration range: 126.0 mg L-1-2697 mg L-1; standard deviation: 646.3 mg L-1) were measured by a hyperspectral camera (wavelength range: 400 nm-1000 nm; resolution: 9 nm). The nitrate concentration of the calluses was measured by the RQ-Flex method. Mathematical models to estimate the nitrate concentration in the calluses from the absorption spectra were developed using the principal component regression (PCR) and partial least squares (PLS) methods, and the correlation coefficient between the measured and estimated nitrate concentration was found to be 0.7363. - The purpose of this research is to clarify the effects of light quality on growth and nitrate concentration of lettuce. The light quality was defined by R/B ratio, which was ratio of photon flux density of red light to that of blue one. Lettuces were cultivated in a growth chamber with 12 h/12 h of light/dark periods under 6 kinds of R/B ratio as follows; 0, 1, 5, 10, 20 and ∞ (all red). The sum of red and blue photon flux density was kept constant as 230 μmol m-2 s-1 at each R/B ratio. In the cultivation 3 kinds of air temperature (20, 22 and 25 degrees centigrade) and 3 kinds of cultivation term (1, 2 and 3 weeks) were considered as experimental treatments. After the cultivation fresh weight, percentage dry matter and nitrate concentration were measured. The fresh weight at 5, 10 and 20 of R/B ratio was larger than that at other R/B ratios and it was found that the larger the percentage dry matter was, the lower the nitrate concentration was. The correlation coefficient between the percentage dry matter and the nitrate concentration became -0.816. The lowest nitrate concentration and the largest percentage dry matter were realized when the R/B ratio was 1. The results of analysis variance did not show the significant difference among 1, 5, 10 of R/B ratio in relation to the percentage dry matter or the nitrate concentration. - In order to understand responses of the halophyte sea aster (Aster tripolium L.) to salt stress at the molecular level, we used a differential screening strategy. Nine cDNAs of genes induced by salt stress were cloned by this method from sea aster. The clones were collectively named SASR (sea aster stress responsive). These cDNAs were mainly associated with osmoprotectant synthesis, protein metabolism, protein degradation or signal transduction. - 日本学術振興会, 科学研究費助成事業, 基盤研究(B), 神戸大学, 01 Apr. 2021 - 31 Mar. 2024スピーキングプラントアプローチによるサフラン子球の生育制御法解明温度条件が異なる三試験区でサフラン子球育成実験を行った。温度条件を一定に保つ低温区に加え温度を栽培前半から段階的に上昇させる早期昇温区と栽培後半から昇温する晩期昇温区を設けた。早期昇温区および低温区では2週間毎、晩期昇温区では毎週4株を収穫し子球のスペクトルを測定した。ハロゲンランプを光源に用いた。526 nm ~ 920 nmの波長帯の分光器 (波長分解能2 nm)および、920 nm~ 1640 nmの波長帯の分光器(波長分解能3 nm)にて分光した。また子球の赤道上の3点から得たスペクトルを平均した後二次微分等の前処理を行い予測モデル構築に使用した。吸光スペクトルを取得後、凍結乾燥を行った球茎試料にメタノールを添加し摩砕試料から糖抽出を行った。得られた抽出液からフルクトース濃度、グルコース濃度およびスクロース濃度をHPLCにて測定した。前処理を行ったスペクトルを説明変量、子球重量または各糖濃度を説明変量として主成分回帰分析にて予測モデル構築を行った。 フルクトース濃度は肥大に伴って増加傾向が、グルコース濃度およスクロース濃度では減少傾向がみられた。多品種の先行研究で確認された肥大に伴うヘキソース・スクロース比の低下が本研究では確認されなかった。このことから改めてシンク強度の指標となり得るパラメーターに関して解析が必要となった。非破壊計測検量線の推定精度(実測値と推定値との間の相関係数)はフルクトースで0.74、グルコースで0.72、スクロースで0.82となった。波長範囲を広げることにより精度向上が期待できる。子球重量については相関係数は0.91となった。これら非破壊計測の結果からシンク強度の新指標を定義した。この指標値は葉の枯れ始めを境に減少に転じる傾向が見られ、非破壊測定可能なシンク強度の新指標となり得ると考えられた。

We have previously shown that lettuce DREB transcription factors activate transcription of target genes involved in stress response and tolerance. However, both the regulons and the paralogs of LsDREB have not been analyzed deeply. In this study, we analyzed the paralogs of the LsDREB gene and the cis-element of the regulon involved in the stress response of lettuce. As a result, it was concluded that a functional LsDREB7 of the wild-type allele under the control of LsDHN12 promoter should be used to enhance stress tolerance in lettuce.

The tested plants were komatsuna. Absorption spectra at each pixel on leaf surface were measured by using a hpyerspectral imaging system. In order to derive mathematical models to estimate the nitrate concentration from absorption spectra, mean spectrum over pixels that were included in small leaf bit was measured. Nitrate concentration of the leaf bit was measured by HPLC. This measurement was applied to 459 leaf bits that included leaf blades and midribs. The mathematical models were developed for leaf blade or midrib separately. Correlation coefficient between the measured and estimated concentration of evaluation data showed more than 0. 91 in both the models. These models were applied to visualize the distribution image of nitrate concentration in a leaf. Then sections of high or low concentration were detected and were cut off from the leaf. Variations of NR gene expression in the leaf sections were measured by PT-PCR and agarose gel electrophoresis methods. Results of this analysis showed that there was not clear correlation between the nitrate concentration and variation of NR gene expression. However, variations of NR gene expression tended to increase with increment of nitrate concentration from initial concentration. It was clarified that the increment of nitrate concentration was considered as one of the factors to induce NR gene expression.Competitive research funding

In this study, an efficient method was established to evaluate lettuce resistance to tipburn in vitro. A sensitive cultivar identified by this method was validated its utility as indicator plants to monitor environmental conditions. On the other hand, to identify the responsible gene for tipburn incidence, four cDNAs corresponding to cation/H^+(CAX) exchanger were isolated from lettuce(LsCAX2, 3a, 3b, and 5). Sequence alignment revealed that both A and B domains which involved in calcium selection were conserved in two LSCAXs. The different expression of LsCAX genes by CaCl_2 between sensitive and resistant cultivars suggested the relevance of these genes to tipburn incidence. AtDREB1A was supposed to be another responsible gene. Overexpression of AtDREB1A in lettuce resulted in increasing tip-burned leaves. This result indicated that tipburn incidence is involved in stress response of lettuce.Competitive research funding

Then an optimum condition to reduce the nitrate concentration has been searched. Accumulation of nitrate in leaves depends on a balance the nitrate uptake from root against the nitrate reduction by nitrate re-ductase (NR). The activation of NR by light is known well. However effect of light quality on NR activation is not analyzed in detail. The followings were focused in this research. 1. Effect of light quality and air temperature on nitrate concentration in lettuce leaves. The light quality was defined by R-B ratio that was ratio of photon flux density of red light to that of blue light. Light source was LED. 2. Effect of light quality on nutrition uptake. The followings were clarified in this research. 1. The optimum conditions of R-B ratio and air temperature were 10 or 5 and 20℃ to keep low nitrate concentration, high percentage dry matter and good appearance. 2. Characteristics of nutrition uptake was not affected by R-B ratio. 3. Non-destructive measurement method was developed by using near infrared spectroscopy. Finally, a regression equation was derived from this method. The coefficient of determination between the measured and estimated nitrate concentration was 0.7809. Substituting the measured near infrared spectra for the regression equation, time course of nitrate concentration in lettuce leaves was measured during cultivation experiment. Comparing the measured concentration by HPLC with the estimation, some sample showed large error. It is necessary to improve the estimation accuracy of the regression equation.Competitive research funding
- 日本学術振興会, 科学研究費助成事業, 奨励研究(A), 神戸大学, 2001 - 2001アクアポリン遺伝子を用いた耐乾燥性・耐塩性・耐冷性カリフラワーの作出平成13年度は,以下の3項目の内容で研究を行い,それぞれの結果が得られた. 1.カリフフワー花蕾培養法および効率的な植物体再生系の確立 平成12年度に行った実験によって確立できた高い再分化率を示すカリフラワーの再分化系をさらに発展させ,「花蕾すりおろし法」の適用について検討した.「花蕾すりおろし法」とは,おろし金などを使用してカリフフワーの花蕾球を細かい断片にして培地に置床する方法であり,操作が簡単で時間も短縮できる.カリフフワーの再分化植物体の獲得において,「花蕾すりおろし法」を用いた再分化系は,通常の花蕾を切除して培地に植える再分化系と比較して再分化効率・順化効率が高いと考えられた. 2.カリフフワー形質転換系の検討 アグロバクテリウムを用いてカリフラワーのすりおろした花蕾から直接感染させたが,形質転換個体の獲得には至っていない.この原因としては,アグロバクテリウムと植物の共存期間・感染時の濃度等が考えられた. 3.ウラギクのsamip遺伝子を導入した植物用形質転換ベクターの構築 平成12年度の結果により全長の塩基配列が明らかになったsamipAおよびsamipB遺伝子を,バイナリベクターpBE2113のT-DNA領域に挿入し,ウラギクのアクアポリンをコードする遺伝子を含む植物用形質転換ベクターを構築した.samip遺伝子が35Sプロモーターの下流にクローニングされていることを,制限酵素消化および塩基配列決定によって確かめた.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), KOBE UNIVERSITY, 1999 - 2000The Efferts of UV-A and UV-B radiation on the expression of genes in flower pigment biosynthesisThe color-changing rose 'Ehigasa' , which petals are redden after flowering under natural daylight, was investigate to clarify the effect of UV light quality (UV-A : 400-320 nm and UV-B : 320-280 nm) on flower pigment (anthocyanin) biosynthesis. Qualification of anthocyanidin skeleton and the bound sugars was investigated by separating of pigments and partial hydrolysates by PC and by measuring absorption spectrum. HPLC analysis of the total anthocyanin content and the ratio of main two anthocyanins to total anthocyanin content at differential developmental stages and light treatment. Main anthocyanins contained in petals of 'Ehigasa' were qualified as cyanidin 3,5-diglucosides (Cy-3G5G) and cyanidin 3-glucoside (Cy-3G). Biosynthesis of anthocyanin stimulated by UV was enhanced with increasing irradiation of visible light at the same time. Tendency of increasing ratio of Cy-3G was observed according to developing flower bud, and the quantity of Cy-3G was exceeded that of Cy-3G5G at unfolded. Leaf lettuce (cv. Red fire) which has colorful red pigment "anthocyanin" was treated with various UV supplemental irradiations, Chlorophyll and anthocyanin amount was much increased by UV irradiation with all wavelength. UV-B(>290 nm) radiation had no effect on the plant growth and the UV-B(>280 nm) affected leaf growth destructively. Even the low irradiance of UV-B280 was fatal. When the supplemental UV irradiation practiced at night only, the growth and development inhibited largely. While day and night irradiation result in no change in leaf color (the amount of pigments did not increased) but has no effect on growth.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), KOBE UNIVERSITY, 1999 - 2000Physiological and ecological mechanisms of response and adaptation of horticultural plant resources to compound environmental stressIn various serious environmental stresses, we investigated mechanism of response and adaptation of the horticultural plant resources to ultraviolet-B radiation and salt stresses. 1. By changing the various irradiation conditions such as exposure time, strength, and wavdength regions of UV-B, what kind of failure was generant to the plant by the UV-B irradiation, the physiological and ecological responses of five vegetable plant species to UV stress were investigated. When UV-B, which reaches the surface, was divided into shorter wavelength and longer wavelength regions than 300 nm, UV of 300nm or less was not related to increase and decrease of visible light, and it showed the almost fixed quantity. For cabbage, spinach, tomato, eggplant, and radish, UV-B was irradiated in the dark period, or light period, and the results was compared with the control. High-intense UV-B in the dark period made the surface of tested vegetable leaves into the bronze, and the leaf margin curled inside, and contracted. Visible light showed the effect that eases the damage by the UV-B irradiation. Tomato had the most UV-B sensitivity 280〜290nm of UV-B gave very intensive negative effects to the vegetables tested. Leaves that UV-B was irradiated had lacked the flexibility, and also decreased on the water content. The effect on the growth was strongly observed in younger leaves. 2. Physiological response (growth, absorption of ions and water physiology) and photosynthesis in the vegetative growth stage of Aster tripolium L.Which was able to grow under the hyper saline environment, were investigated in order to clarify the salt tolerance mechanism. The growth lowered by water stress, and NaCl stress (300mM), and it decreased more in water stress than NaCl stress. Absorption of Na^+ and Cl^+ ino the leaves was higher in the NaCl stress than water stress, and the Na+/K+ ratio was also higher Water potential of the leaves in control, water stress, and NaCl stress lowered in order. Turgor pressure was almost same in control and NaCl stress, and lowered in water stress. Under the atmosphere CO_2 condition (350 ppm) the rates of stomatal limiting of photosynthesis in control, water stress, and salt stress were 33%, 52% and 44% of control respectively Without the stomatal limiting the light saturation photosynthetic rate was decreased in the water stress, but not in the NaCl stress. The photosynthetic rate was decreased because of limiting the supply of CO_2 through the partial closure of the stoma under NaCl stress. Light and dark reaction systems of photosynthesis, however, were protected in NaCl stress. In the meantime, the photosynthetic rate in water stress was lowered by closure of the stoma and decrease in the ability of the dark reaction system.
