- Current treatment options for osteoporosis primarily involve pharmacotherapies, but they are often accompanied by undesirable side effects. Utilization of mechanical stress which can noninvasively induce bone formation has been suggested as an alternative to conventional treatments. Here, we examined the efficacy of mechanical stress induced by electrical stimulation, radial extracorporeal shock waves, and ultrasound for estrogen-deficient osteoporosis. Female Wistar rats were divided into following five groups: sham-operated group, untreated after ovariectomy, and treated with electrical stimulation, radial extracorporeal shock wave, or ultrasound starting at 8 weeks after ovariectomy for 4 weeks. Trabecular bone architecture of the femur was assessed by micro-CT and its biomechanical properties were obtained by mechanical testing. The femurs were further evaluated by histochemical, immunohistochemical, and real-time PCR analyses. Radial extracorporeal shock wave and ultrasound treatment improved trabecular bone microarchitecture and bone strength in osteoporotic rats, but not electrical stimulation. The shock wave decreased osteoclast activity and RANKL expression. The exposure of ultrasound increased osteoblast activity and β-catenin-positive cells, and they decreased sclerostin-positive osteocytes. These findings suggest that mechanical stress induced by radial extracorporeal shock wave and ultrasound can improve estrogen-deficient bone loss and bone fragility through promoted bone formation or attenuated bone resorption.Aug. 2021, Calcified tissue international, 109(2) (2), 215 - 229, English, International magazine[Refereed]Scientific journal

- Fractures associated with osteoporosis are a major public health concern. Current treatments for fractures are limited to surgery or fixation, leading to long-term bedrest, which is linked to increased mortality. Alternatively, utilization of physical agents has been suggested as a promising therapeutic approach for fractures. Here, we examined the effects of ultrasound, radial extracorporeal shock waves, and electrical stimulation on normal or osteoporotic fracture healing. Femoral bone defects were created in normal or ovariectomized rats. Rats were divided into four groups: untreated, and treated with ultrasound, shock waves, or electrical stimulation after surgery. Samples were collected at 2 or 4 weeks after surgery, and the healing process was evaluated with micro-CT, histological, and immunohistochemical analyses. Ultrasound at intensities of 0.5 and 1.0 W/cm2 , but not 0.05 W/cm2 , accelerated new bone formation. Shock wave exposure also increased newly formed bone, but formed abnormal periosteal callus around the defect site. Conversely, electrical stimulation did not affect the healing process. Ultrasound exposure increased osteoblast activity and cell proliferation and decreased sclerostin-positive osteocytes. - OBJECTIVE: Joint contractures are a major complication following joint immobilization. However, no fully effective treatment has yet been found. Recently, carbon dioxide (CO2) therapy was developed and verified this therapeutic application in various disorders. We aimed to verify the efficacy of transcutaneous CO2 therapy for immobilization-induced joint contracture. METHOD: Twenty-two Wistar rats were randomly assigned to three groups: caged control, those untreated after joint immobilization, and those treated after joint immobilization. The rats were treated with CO2 for 20 min once a daily either during immobilization, (prevention) or during remobilization after immobilization (treatment). Knee extension motion was measured with a goniometer, and the muscular and articular factors responsible for contractures were calculated. We evaluated muscle fibrosis, fibrosis-related genes (collagen Type 1α1 and TGF-β1) in muscles, synovial intima's length, and fibrosis-related proteins (Type I collagen and TGF-β1) in the joint capsules. RESULTS: CO2 therapy for prevention and treatment improved the knee extension motion. Muscular and articular factors decreased in rats of the treatment group. The muscular fibrosis of treated rats decreased in the treatment group. Although CO2 therapy did not repress the increased expression of collagen Type 1α1, the therapy decreased the expression of TGF-β1 in the treatment group. CO2 therapy for treatment improved the shortening of the synovial membrane after immobilization and decreased the immunolabeling of TGF-β1 in the joint capsules. CONCLUSIONS: CO2 therapy may prevent and treat contractures after joint immobilization, and appears to be more effective as a treatment strategy for the deterioration of contractures during remobilization.Corresponding, Dec. 2020, Physical Therapy Research, 23(2) (2), 113 - 122, English, Domestic magazine[Refereed]Scientific journal

- STUDY DESIGN: Experimental animal study. OBJECTIVES: Spastic hypertonia is originally believed to cause contractures from clinical observations. Botulinum toxin is effective for the treatment of spasticity and is widely used in patients who have joints with contractures. Using an established rat model with knee contractures after spinal cord injuries, we aimed to verify whether hypertonia contributes to contracture development, and the botulinum toxin improves structural changes in muscles and joint components responsible for contractures. SETTING: University laboratory in Japan. METHODS: To evaluate the effect of hypertonia on contracture development, the rats received botulinum toxin injections after spinal cord injuries. Knee extension motion was measured with a goniometer applying a standardized torque under anesthesia, and the contribution by muscle or non-muscle structures to contractures were calculated by measuring joint motion before and after the myotomies. We quantitatively measured the muscle atrophy, muscle fibrosis, and synovial intima length. RESULTS: Botulinum toxin injections significantly improved contractures, whereas did not completely prevent contracture development. Botulinum toxin was effective in improving the muscular factor, but little difference in the articular factor. Spinal cord injuries induced muscle atrophy, and botulinum toxin significantly accelerated muscle atrophy and fibrosis. The synovial intima length decreased significantly after spinal cord injuries, and botulinum toxin did not improve this shortening. CONCLUSIONS: This animal study provides new evidence that hypertonia is not the sole cause rather is the partial contributor of contractures after spinal cord injuries. - BACKGROUND: Joint contractures are a major complication in patients with spinal cord injuries. Positioning, stretching, and physical therapy are advocated to prevent and treat contractures; however, many patients still develop them. Joint motion (exercise) is crucial to correct contractures. Transcutaneous carbon dioxide (CO2) therapy was developed recently, and its effect is similar to that of exercise. This therapy may be an alternative or complementary approach to exercise. QUESTION/PURPOSES: Using an established model of spinal cord injury in rats with knee flexion contractures, we sought to clarify whether transcutaneous CO2 altered (1) contracture, as measured by ROM; (2) muscular and articular factors contributing to the loss of ROM; (3) fibrosis and fibrosis-related gene expression in muscle; and (4) the morphology of and fibrosis-related protein expression in the joint capsule. METHODS: Thirty-six Wistar rats were divided into three equal groups: caged control, those untreated after spinal cord injury, and those treated with CO2 after spinal cord injury. The rats were treated with CO2 from either the first day (prevention) or 15th day (treatment) after spinal cord injury for 2 or 4 weeks. The hindlimbs of rats in the treated group were exposed to CO2 gas for 20 minutes once daily. Knee extension ROM was measured with a goniometer and was measured again after myotomy. We calculated the muscular and articular factors responsible for contractures by subtracting the post-myotomy ROM from that before myotomy. We also quantified histologic muscle fibrosis and evaluated fibrosis-related genes (collagen Type 1, α1 and transforming growth factor beta) in the biceps femoris muscle with real-time polymerase chain reaction. The synovial intima's length was measured, and the distribution of fibrosis-related proteins (Type I collagen and transforming growth factor beta) in the joint capsule was observed with immunohistochemistry. Knee flexion contractures developed in rats after spinal cord injuries at all timepoints. RESULTS: CO2 therapy improved limited-extension ROM in the prevention group at 2 weeks (22° ± 2°) and 4 weeks (29° ± 1°) and in the treatment group at 2 weeks (31° ± 1°) compared with untreated rats after spinal cord injuries (35° ± 2°, mean difference, 13°; 39° ± 1°, mean difference, 9°; and 38° ± 1°, mean difference, 7°, respectively) (95% CI, 10.50-14.86, 8.10-10.19, and 4.73-9.01, respectively; all p < 0.001). Muscular factors decreased in treated rats in the prevention group at 2 weeks (8° ± 2°) and 4 weeks (14°± 1°) and in the treatment group at 2 weeks (14 ± 1°) compared with untreated rats (15° ± 1°, 4.85-9.42; 16° ± 1°, 1.24-3.86; and 17° ± 2°, 1.16-5.34, respectively; all p < 0.05). The therapy improved articular factors in the prevention group at 2 weeks (4° ± 1°) and 4 weeks (6° ± 1°) and in the treatment group at 2 weeks (8° ± 1°) compared with untreated rats (10° ± 1°, 4.05-7.05; 12° ± 1°, 5.18-8.02; and 11° ± 2°, 1.73-5.50, respectively; all p < 0.05). CO2 therapy decreased muscle fibrosis in the prevention group at 2 weeks (p < 0.001). The expression of collagen Type 1, α1 mRNA in the biceps femoris decreased in treated rats in the prevention group at 2 and 4 weeks compared with untreated rat (p = 0.002 and p = 0.008, respectively), although there was little difference in the expression of transforming growth factor beta (p > 0.05). CO2 therapy did not improve shortening of the synovial intima at all timepoints (all p > 0.05). CO2 therapy decreased transforming growth factor beta immunolabeling in joint capsules in the rats in the prevention group at 2 weeks. The staining intensity and Type I collagen pattern showed no differences among all groups at all timepoints. CONCLUSION: CO2 therapy may be useful for preventing and treating contractures after spinal cord injuries. CO2 therapy particularly appears to be more effective as a prevention and treatment strategy in early-stage contractures before irreversible degeneration occurs, as shown in a rat model. - Joint mobility decreases in the elderly and in diabetics, this process is thought to be caused by accumulation of advanced-glycation end products (AGEs). Here, we aimed to elucidate the role of AGEs in joint contracture formation in rat knees. Rats were injected with ribose or saline into the knees twice weekly for 8 weeks. Pentosidine (AGE) levels were measured in the knee-joint tissues. After serial injections, rats were subjected to unilateral knee-joint immobilization in a flexion position for various periods. At day 21, the passive knee ranges of motions (ROMs) were measured. Knee joint histopathology were assessed, and the expression of fibrotic genes in the posterior joint capsules was examined using real-time PCR. Ribose injection induced a 7.0-fold increase in pentosidine levels relative to saline injection. Joint immobilization resulted in equal myogenic ROM restriction in both groups. Arthrogenic ROM restriction was greater with ribose injection in the immobilized joints (p < 0.05), but was not affected in nonimmobilized joints. Type-I (COL1A1) and type-III (COL3A1) collagen gene expression increased significantly in immobilized joints relative to nonimmobilized joints in the ribose group, but was not affected in the saline group. Ribose injection increased COL1A1 expression slightly and COL3A1 expression significantly in immobilized joints. Histologically, inflammatory changes appeared at day 3 of immobilization and peaked at day 7. These responses trended to be more severe and prolonged in the ribose group than in the saline group. Our data provide evidence for a causal relationship between AGEs and joint contracture formation following immobilization. © 2017 Orthopaedic Research Society. Published by Wiley Periodicals, Inc. - Objective : We performed a systematic review to investigate improvements in body function and structure, activity, and health-related quality of life from exercise programs of >150 days' duration in patients with knee osteoarthritis (OA). Methods : We collected data on randomized controlled trials (RCTs) comparing the effects of exercise intervention with those of either nonintervention or psychoeducational intervention in patients with knee OA. Data were collected on pain, stiffness, muscle strength, range of motion, flexibility, maximal oxygen uptake, proprioception, time to climb a flight of stairs, disability as assessed by the Western Ontario and McMaster Universities Arthritis Index (WOMAC), walking distance (e.g., 6-min walking-distance test), walking time (e.g., time to walk arbitrary distances), walking velocity, and health-related quality of life assessed using the SF-36® Health Survey (SF-36). The Grading of Recommendations Assessment, Development and Evaluation (GRADE) system was used to determine the quality of the evidence. Results: Forty-one RCTs were identified. Outcomes that were significantly affected by exercise programs of >150 days were knee flexion muscle strength at a functional level, time to climb a flight of stairs and walking distance at activity level, and SF-36 General Health, Physical Functioning, Role-Physical, and Bodily Pain scales. The quality of evidence for these outcomes was moderate to high. - Denervation alters the metabolism of the extracellular matrix (ECM) in skeletal muscle; however, the underlying mechanisms of ECM remodeling are not fully understood. The aim of this study was to elucidate the dynamic features of the ECM regulatory process in the early phase of denervated skeletal muscle in male Wistar rats. We investigated the expression of collagens (total, type I, and type III), transforming growth factor beta 1 (TGF-β1), and matrix metalloproteases (MMPs) together with their endogenous inhibitors (TIMPs), at the mRNA and/or protein level in the soleus muscles of control animals and at days 3, 7, and 14 post-denervation. Expression of mRNA encoding collagens was decreased at days 3 and 7, and had recovered by day 14, in parallel with total collagen protein content. Content of TGF-β1 protein was elevated sequentially, up to a maximum of 158% at day 14 post-denervation (P < 0.05), as was TIMP-2 mRNA expression (272% at day 14), whereas MMP-1, MMP-2, and TIMP-1 mRNA expression was not affected at any stage. The initial reduction of collagen mRNA may be responsible for hypoactivity caused by the disappearance of contractile function. - 加齢により関節軟骨は退行性変化を呈し,主要構成要素であるプロテオグリカン含量やⅡ型コラーゲンの減少が認められる。この関節軟骨の退行性変化は運動がもたらす関節への機械的刺激によって抑制および修復させることが報告されている。本研究では老齢ラットモデルの距腿関節軟骨を組織学的に分析し,走行運動およびバランス運動が距腿関節軟骨にあたえる影響を検討した。結果,老齢ラット群の関節軟骨は若齢ラット群と比較してⅡ型コラーゲン及び関節軟骨厚が減少し,関節軟骨表層部に亀裂が認められた。一方で,老齢ラット通常飼育群に比較してバランス運動群の軟骨厚が増加していた。このことから加齢によって距腿関節軟骨退行性変化を呈するが,関節運動を伴う機械的刺激によって関節軟骨変性を抑制,改善に作用する可能性があることが示唆された。Corresponding, 公益社団法人 埼玉県理学療法士会, Apr. 2011, 理学療法-臨床・研究・教育, 18(1) (1), 61 - 66, Japanese[Refereed]Scientific journal

- ラット脊髄損傷後早期の関節拘縮進行過程における筋と関節構成体の関与Introduction: The objective of this study was to determine the rate of contracture progression and the direction of loss in joint movement, and to identify the relationship between the muscular and articular factors leading to contractures in acute spinal cord injuries. Methods: Twenty female Wistar rats were allocated to one of 5 equal sized groups that were assessed respectively at preinjury, 3, 5, 7, or 14 days after spinal cord injuries. The degree of contractures was assessed by measuring the femorotibial angle on both hindlimbs with the use of a goniometer. Knee joint motion was measured for flexion and extension. Myotomy of the transarticular muscles was then performed and range of motion was measured again. Results: The spinal cord-injured animals demonstrated a flaccid paralysis during the first 7 days postinjury and thereafter spastic paralysis. Contractures progressed for 14 days after spinal cord injuries, regardless of changes in muscle tone. Loss in joint movements was produced almost exclusively by a loss in the extension range of motion. Both the muscles and the articular structures contributed to the progression of the contracture, and these components are factors in the promotion of contracture development. In particular, the muscular factors were greater than the articular factors in extension angular displacement in which the range of motion was restricted. - Motor deficits after spinal cord injury arise from damages to the descending spinal pathways and ventral motoneurons (VMN). In contrast to data on damages to the white matter or the descending spinal pathways, few quantitative data on damages to VMN after injury are available currently. The purpose of this study was to examine quantitatively the temporal and spatial pattern of VMN loss after spinal cord compression. Two groups of adult female Wistar rats were used in this study: rats which were subjected to spinal cord compression in short duration with an aneurysm clip (experimental group) and rats which were subjected to a sham-operation (control group). Using serial cross-sections of the spinal cord, VMN were counted up to the 7th day after surgical intervention at 0, 1, 2, and 3 mm rostral and caudal to the lesion epicenter (experimental group) or to the median of the serial sections (control group). At 15 minutes after the compression, VMN were lost only at the epicenter section and no VMN were observed there. By 8 hours, VMN loss had spread to next 1 mm rostral and caudal section to the epicenter. Virtually, no further loss was detected between 8 hours and later time points. This study showed that compression to the adult rat spinal cord in short duration led to VMN loss, which progressed acutely and expanded modestly. - 頚髄損傷の評価では,頚髄の損傷の程度と損傷高位が重要である.米国脊髄損傷協会は,脊髄損傷の障害の評価法を発表し,脊髄損傷の神経学的および機能的分類のための国際基準として現在国際的に使用されている.しかし可能性に挑戦するリハビリテーションとしては,より詳細な高位分類が必要である.Zancolli分類は頚髄損傷四肢麻痺の上肢機能を細かく分類し,リハビリテーションからみても車椅子ADLが自立する可能性のあるC6を細かく分けているのは有用である.しかしマット上基本動作,移乗・移動などの動作が自立するか否かの判断に重要な肩甲帯筋群の評価がない.従来肘伸展筋である上腕三頭筋はC7髄節筋であるが,Zancolli分類ではC6髄節残存群のサブグループとしているのは混乱をまねく.そこでZancolli分類を改良し,損傷高位別の機能到達目標を決定するための評価表を作成し,ADLが自立する可能性について検討した.改良Zancolli分類でみるとC6BⅡが車椅子ADL自立の境界レベルである.広島大学大学院保健学研究科, Aug. 2001, Journal of health sciences, Hiroshima University, 1(1) (1), 73 - 77, Japanese[Refereed]Research institution

- Human T-Lymphotrophic virus Type-I (HTLV-I) associated myelopathy (HAM/TSP), which manifests as paraplegia is a common disease in Kagoshima prefecture. For planning the rehabilitation of these patients, we determined their muscular power of truncal flexion and extension as posture-supporting muscles in eccentric and concentric contraction with KINCOM 5000, and compared the results with those of healthy controls. Muscular strength of truncal extension in those with HAM was significantly decreased; however, that of truncal flexion was not decreased when it was compared to that of healthy controls. These results indicate that the decreased muscular power of truncal extension in patients with HAM is a characteristic. The mechanism is still unknown. Patients with HAM usually use canes and support their body weight with the non-impaired upper extremities, which may have resulted in the sparing of the muscular power of truncal flexion. - Investigating joint contractures following central nervous system injury : through comparison with contractures following joint immobilization近年,動物モデルの分析を通じて,関節固定そして中枢神経障害にともなう関節拘縮はそれぞれ区別されるべきとの根拠が提示されるようになってきた。これまでに得られた結果は(1)原因が異なると,関節拘縮の進行に筋と関節構成体のそれぞれが関与する割合は異なる,(2)滑膜に生じる病理変化は中枢神経障害モデルの変化の方がより多面的である,(3)原因が異なると,関節軟骨の厚さに異なる変化が異なる分布で発生する,と要約できる。現時点では治療法は区別されていないが,こうした知見は原因別に最適化された治療アプローチが必要であることを示している。これまで各種治療の関節可動域の治療効果は,主に関節固定モデルを用いて検証されてきた。そこで得られた情報は,中枢神経障害における関節拘縮の治療でも利用できる可能性がある。しかし,病態は同一ではないため,治療による可動域,そして組織への効果は,原因ごとに検討される必要がある。Analyzing experimental animal tests, there is growing evidence demonstrating that respective contractures following joint immobilization and central nervous system(CNS)injury should be distinguished. Results from the tests could be summarized as follows:(1)roles of muscular and articular factors in the progression of contractures are different causedependently, (2)pathological changes in synovial membrane are more complex in contractures following CNS injury,(3)depending on the cause of contractures, differences in articular cartilage thickness appear in different manners and locations. These findings indicate that differing optimized therapeutic approaches are required cause-dependently, whereas such a distinction is not made at present. To date, the effect of therapeutic strategies on joint range of motion has been studied using experimental joint immobilization models mainly. Information from these studies could be used to treat contractures following CNS injury. - Examining the effect of muscle stretching and self-training (hospital-prescribed method) for chronic lower-back pain. The maximum treatment period in the survey was 3 months since the previous outpatient consultation. The test group was divided into three: a control group, Exercise 1 group (carrying out SLR with trunk muscle contract relaxation), and Exercise 2 group (carrying out SLR with trunk muscle strengthening and lower back stretching).The Japanese Society of Lumbar Spine Disorders, Oct. 2009, The Journal of Japanese Society of Lumber Spine Disorders, 15(1) (1), 45 - 51, Japanese
For each group, the pain, change of physical function and health-related QOL were examined at 3 months and 6 months after the start of the research period.
The results showed that in the Exercise 2 group, pain and physical function was improved at 3 months; in the Exercise 1 group, almost the same effect was shown as in the Exercise 2 group at 6 months. The item which was most closely related to pain alleviation was trunk flexibility and trunk extension strength.

- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Hiroshima International University, 01 Apr. 2021 - 31 Mar. 2026Relationships between femoral morphological abnormalities / joint malalignment and locomotor disorders本年度の研究では,筋骨格が未成熟なラットを用い,免荷による後肢へのメカニカルストレスの減少が,1)大腿骨形態,2)膝関節アライメント,3)膝関節機械的特性に及ぼす影響を調査した.4週齢の雌性ラットに後肢懸垂を2,4,8週間実施し,以下の実験を行った. 1)では,X線CT撮影画像を用い,大腿骨幾何学形態への影響を調査した.後肢懸垂により,大腿骨近位では,頚部前捻角(Anteversion angle: AVA)が増加した.大腿骨遠位では,外側顆部(Lateral condyle: LC)では成長が抑制された一方で,内側顆部(medial condyle: MC)には成長に影響を及ぼさなかった.滑車角(Trochlear angle : TA)は,成長に伴い生じる外方から内方への傾斜が,後肢懸垂により抑制された.AVA,LC,MC/LC, TAの間には強い相関が認められた. 2)では,膝関節アライメントに及ぼす影響を調査した.後肢懸垂により,大腿骨に対する膝蓋骨の傾斜角度(Patellar Tilt Angle)は外方に傾斜し,脛骨粗面-滑車溝間距離(TT-TG)により脛骨の外旋が示された.加えて,膝蓋骨の側方変位の指標であるBisect offsetは,膝蓋骨の外方への移動を示した. 3)では,膝関節全体および前十字靭帯(Anterior Cruciate Ligament: ACL)における機械的特性への影響を調査した.万能試験機を用い,後肢懸垂後の膝関節弛緩性(Joint laxity),ACLのstiffnessおよび破断強度を調査した.その結果,4-8週間の後肢懸垂により,いずれの機械的強度の指標も変化し,関節弛緩性が出現することが示された.

However, no effective treatment has been developed so far. In the present study, we aimed to develop a novel therapeutic approach for joint contractures using transcutaneous carbon dioxide absorption. Our findings indicate that transcutaneous carbon dioxide may improve knee range of motion after injuries central nervous system or joint immobilization. We also revealed that transcutaneous carbon dioxide decreases muscular and articular fibrosis, thereby improving knee joint contracture.

First, a combination of AGEs accumulation and treadmill running was tested. AGEs accumulated knee joint was not induced any pathological changes. Second, we tested the effects of AGEs on joint mobility. AGEs accumulation in joint components did not affect joint mobility. However, joint contracture following immobilization was aggravated in AGEs accumulating joints with concomitant upregulation of collagen genes and protein in joint capsule. This study is the first to elucidate the relationship between AGEs and joint contracture formation following immobilization via enhancement of fibrosis in joint capsule.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Kio University, 01 Apr. 2012 - 31 Mar. 2016Effects of treadmill exercise on respiratory function, diaphragm muscular function and respiratory center in rats with emphysema.The purpose of this study was to determine the effects of treadmill exercise on respiratory function and skeletal muscle in rats with emphysema. Wistar rats were randomly divided into three groups: the sham (SH) group, pulmonary emphysema (PE) group, and emphysema + exercise (EX) group. Cigarette smoke solution and lipopolysaccharide were intratracheally administered for 4 weeks in the PE and EX groups. The rats in the EX group were made to run on treadmills in the latter 2 weeks of the experiment. Many macrophages were observed in the lung tissue of the PE group. In the EX group, however, the population of macrophages was smaller and the specific force of the diaphragm and EDL muscles was higher than in the PE group. Moreover, the degree of inflammation in the pulmonary tissue was reduced in the EX group. These results suggest that adaptive exercise may improve not only respiratory and muscle functions but also inflammation of the pulmonary tissue associated with emphysema.

In this study, whether it is possible to activate the nervous system that has been decreased by aging and to verify the effect of the running and balance exercise. Activation of the motor neurons stained of the lumbar spinal cord was less change in some exercise. Their expression of neurotrophins, neurogenesis and neurpeptide were enhanced and the activity of apotosis wad down-regulated by locomotor exercise in the spinal cord of the difference of age. Improvements due to the motion, not the nerve itself, it is necessary to consider in the pleiotropic functions for related factors activated neural activity.

First, we erected a method that couples microbeads with blood cells. Secondly, we glued a magnet to the surface of rat’s skull and injected the microbead - blood cell complexes into the rat’s internal carotid artery. As a result, we succeeded to occlude the rat’s middle cerebral artery. To lessen the damage to rats and to make the surgical technique simple, we conducted next experiment under another condition. The experiment showed that the middle cerebral artery of a rat was not occluded in that condition.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Saitama Prefectural University, 2009 - 2011Elucidation of the natural healing mechanism after knee anterior cruciate ligament tear and it's clinical applicationThe natural healing of the ruptured anterior cruciate ligament(ACL) using a new animal model was observed by mechanical, histological and immunohistochemistrical methods. The ACL was cut completely by the mini-scissors and then the knee joint was controlled normally with artificial ligament. The ruptured ACL was healed in all rats. Eight weeks after ACL cutting, the Type III collagen was increased in the healing ligament. After 8 weeks and 40 weeks after the ACL cut, the healing ligament was half strength compared to the normal ligament. The expression of MMP-1, MMP-13 and TIMP-1 in medial and lateral meniscus and rupture ACL was increased after ACL cut. Expression of MMP-13 in the joint control group has been suppressed compared to the no joint control group. It is suggested that abnormal joint movement under the gravity is increased to express of proteolytic enzymes of both meniscus and rupture ACL in the knee joint.

Exercised rats showed a quantitative increase neurotrophin immunostaining that was mostly evident in the spinal cords. In biochemical assay, exercise increases selectively expression of neurotrophins and receptors mRNA in the lumber spinal cord in senescence. These results suggest that exercise enhanced neurotrophins and their receptors by physiological process that it is endogenous in the lumber spinal cords which innervate the knee joints.

After the operation, the tidal volume of rats with C2 level injury decreased about 5%, and that of rats with C3 level about 25%. The tidal volume of rats with C5 to C7 levels has decreased about 75-95%. In contrast, their diaphragm muscle activity has increased about 10-20%. It could bring out that the respiratory function of patients with cervical spinal cord injury not only affects the activity levels of daily living but also has a significant effect on life support.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Saitama Prefectural University, 2007 - 2008Search of natural healing for conservative treatment after anterior cruciate ligament rupture前十字靱帯は完全断裂であっても関節運動の制動と早期から運動を行うことによって, 破断したACL が治癒することを証明するための動物実験モデルを作製することが本研究の主目的であった.後肢懸垂モデル, 関節包外関節制動モデルを考案し, 靱帯の治癒を検証した.後肢懸垂モデルでは索状の瘢痕組織が観察されたが, 荷重運動を開始すると瘢痕組織は消失し, 変形性膝関節症が発生した.関節包外関節制動モデルでは肥厚した瘢痕組織が観察されたが, 結果にばらつきが発生し, 関節コントロールが関連すると考えた.
