    Purple yam (Dioscorea alata L.) is a tuber widely distributed in the tropics and subtropics. We previously isolated several acylated anthocyanins from purple yam. In this study, purple yam extract was orally administered to rats and the absorption of the constituent anthocyanins was investigated. Acylated anthocyanins were detected in the plasma, confirming that they were absorbed into the body as their intact forms. The time required to reach the maximum anthocyanin concentration was 15 min, indicating rapid absorption into the blood. In addition, more than half of the total amount of anthocyanins excreted into the urine was excreted within the first 6 h after administration. To the best of our knowledge, this is the first report of the detection of characteristic acylated anthocyanins from purple yam in rat plasma and urine after oral administration.

    BACKGROUND: Low-grade systemic inflammation may be a key player in the immune activation that has been reported for mental health deterioration. We hypothesised that elevated serum levels of inflammatory cytokines increase neuroinflammation and exacerbate depressive symptoms. METHODS: The participants were part of a cohort study for whom data was available for both 2015 and 2019. In 2015, blood samples were collected from 232 participants. Their depressive symptoms were assessed both 2015 and 2019 using the Centre for Epidemiologic Studies Depression Scale (CES-D) (n = 33). The multiplex immunoassay system (Luminex® 200) was used to measure the serum concentrations of IL-6, IL-10, IL-12, IL-17A and TNFα. Data were analysed using linear models with the level of significance considered to be p < 0.05. RESULTS: After controlling for age, BMI, smoking and alcohol consumption, in 2015 the serum concentrations of IL-17A and TNFα in 2015 were significantly positively associated with the CES-D scores of women (standardised β (B) = .027, p < 0.01 and B = 0.26, p < 0.01, respectively). The serum concentrations of IL-17A and TNFα of men were significantly positively associated with the CES-D scores of 2019 (B = 0.62, p = 0.02 and B = 0.59, p = 0.02, respectively). CONCLUSIONS: In this cross-sectional study, we found a significant positive correlation between the depressive symptoms and serum TNFα and IL-17A levels of women. In addition, our longitudinal findings suggest the possibility that TNFα and IL-17A could elevate the depressive symptoms of men.

    To maintain a beneficial concentration of eicosapentaenoic acid (EPA), the efficient conversion of its precursor, α-linolenic acid (α-LA), is important. Here, we studied the conversion of α-LA to EPA using ICR and C57BL/6 mice. A single dose of perilla oil rich-in α-LA or free α-LA had not been converted to EPA 18 h following administration. The α-LA was absorbed into the circulation, and its concentration peaked 6 h after administration, after which it rapidly decreased. In contrast, EPA administration was followed by an increase in circulating EPA concentration, but this did not decrease between 6 and 18 h, indicating that the clearance of EPA is slower than that of α-LA. After ≥1 week perilla oil intake, the circulating EPA concentration was >20 times higher than that of the control group which consumed olive oil, indicating that daily consumption, but not a single dose, of α-LA-rich oil might help preserve the physiologic EPA concentration. The consumption of high concentrations of perilla oil for 4 weeks also increased the hepatic expression of Elovl5, which is involved in fatty acid elongation; however, further studies are needed to characterize the relationship between the expression of this gene and the conversion of α-LA to EPA.

    We previously showed that social stress exposure in mature adult mice increased blood corticosterone concentrations at 2 days, disrupted hepatic lipid metabolism-related pathway at 30 days, and increased the risk of overweight with hepatic hypertrophy at 90 days. To further investigate the effects of aging on the physiological responses to social stress, we conducted a study using male BALB/c mice at the ages of 2 months (mature age), 14 months (middle age) and 26 months (old age), and exposed them to confrontation stress for 2 or 7 days. Blood corticosterone concentrations were increased at 2 days of stress, and then returned to baseline concentrations. This change was observed only at 2 months of age. We further examined the effect of aging on hepatic gene expression of fibroblast growth factor-21 (Fgf21) and found that its expression was significantly decreased after 7 days of stress at 14 months of age and after 2 days of stress at 26 months of age, indicating these decreasing effects became more pronounced with age. In conclusion, our study suggests that hepatic Fgf21 expression decrease under exposure to confrontation stress at middle or more age, indicating that stress response on Fgf21 related pathway might be more pronounced with age when exposed to stress.

    BACKGROUND/AIM: Excessive fructose intake reportedly leads to the development of nonalcoholic fatty liver disease (NAFLD). In our previous study, we reported that plasma activities of alkaline phosphatase (ALP) isozymes were markedly changed in rats with excessive fructose intake-induced hepatomegaly. In this study, we examined ALP isozyme activity prior to the occurrence of hepatomegaly, and investigated the effect of the timing of sample collection, to explore its potential as a biomarker. MATERIALS AND METHODS: After 1-week intake of a 63% high-fructose diet (HFrD), blood samples were collected from male rats during sleep or active phases to analyze biochemical parameters. RESULTS: Body and liver weights were similar between the HFrD and control diet groups, indicating that hepatomegaly due to excessive fructose intake had not occurred. The triglyceride levels and glutamate dehydrogenase (GLDH) activity were significantly elevated to similar degrees at both time points. HFrD intake significantly increased liver-type ALP (L-ALP) activity, stimulating it by 12.7% at the sleep phase and by 124.3% at the active phase. HFrD consumption also significantly decreased intestinal-type ALP (I-ALP) at the active phase, but only showed a decreasing trend during the sleep phase. CONCLUSION: Measurements of plasma ALP isozyme and GLDH activity, and triglyceride levels are effective early biomarkers of impending NAFLD caused by excessive fructose intake. L-ALP and I-ALP activities during the active phase are particularly sensitive for detection of excessive fructose intake before the occurrence of NAFLD.

    BACKGROUND/AIM: The habitual consumption of excessive fructose is associated with the onset and progression of lifestyle-related diseases, such as nonalcoholic fatty liver disease (NAFLD). In this study, we investigated the physiological changes observed when consuming a diet containing excessive fructose on the viewpoints of hepatotoxicity biological markers using a rat model and explored the biomarker candidates that could detect the effects of excessive fructose intake at an early stage. MATERIALS AND METHODS: Male rats were fed 63% high fructose diet (HFrD) ad libitum and their blood samples were collected before and at 1, 2, 3, and 4 weeks after allocation. The plasma biochemical parameters, hepatotoxic enzyme activities including alkaline phosphatase (ALP) isozymes were analyzed. RESULTS: HFrD consumption for 4-weeks created NAFLD-like symptoms, including elevated plasma lipid parameters and hepatotoxicity markers, as well as fat accumulation in the liver compared with rats consuming a control diet. Alanine aminotransferase (ALT) and glutamate dehydrogenase (GLDH) were increased from the 3rd and 2nd weeks, respectively, but no changes were observed on ALP activity. However, the daily consumption of the HFrD increased the plasma activities of liver-type ALP isozyme, and decreased plasma small intestinal-type ALP isozyme soon after the start of feeding. CONCLUSION: ALP isozyme analysis in combination with GLDH and ALT activities in the plasma samples could be a useful tool to detect the physiological changes induced by excessive fructose intake at an early stage of the development of NAFLD.

    Background: Maltose is a disaccharide formed from two units of glucose. The amount of maltose in raw sweet potatoes reportedly increased by more than 40-fold after baking. We aimed to investigate the effects of maltose consumption on diet-induced obesity using a mouse model.Methods: Male C57BL/6J mice were divided into three dietary groups: a control diet of American Institute of Nutrition (AIN)-93 (CD), an AIN-93-based 30% high-fat diet (HFD), or an HFD diet containing 7.0% maltose (HFD+Mal). After 13 weeks, body mass index, blood glucose, lipid parameters, including total cholesterol and triglyceride, and hepatic fatty acid content were evaluated. Results: After 6 weeks on the special diet, weight gain was significantly higher in the HFD+Mal group than in the HFD group. The body mass index rose steadily and was significantly higher in the HFD+Mal group (5.03 ± 0.22), compared to the CD (3.32 ± 0.30) and HFD (4.57 ± 0.40) groups at 12 weeks. The relative liver weight per weight was also significantly higher in the HFD+Mal group than in the other two groups. The increases in blood glucose levels were more significant in the HFD+Mal group compared to the HFD group, as were the plasma levels of total cholesterol, high-density lipoprotein (HDL)-cholesterol and non-HDL-cholesterol. In the liver, levels of the following fatty acids increased in both the HFD and HFD+Mal groups relative to those in the CD group: C14:0 (myristic acid), C16:0 (palmitic acid), C18:0 (stearic acid), C18:1 (oleic acid), C18:2 (linoleic acid) and C18:3 (α-linolenic acid). Additionally, the C16:0 content in the HFD+Mal group was significantly higher than that in the HFD group.  Conclusion: The results of this study suggest that daily consumption of maltose exacerbated the development of obesity and related parameters, including body mass index and plasma cholesterol levels, under the high-fat diet consumption. It is possible that the consumption of maltose-rich cooked sweet potatoes, as part of an overall HFD, might exacerbate HFD-induced overweight.Keywords: high-fat diet-induced obesity, maltose, mice, roasting, sweet potato

    In recent years, the incidence of type I hypersensitivity including hay fever has been increasing year by year in Japan. Our previous study using mice showed that only oral, but not intraperitoneal, administration of fucoidan extracted from seaweed (Saccharina japonica) suppressed type I hypersensitivity by secretion of galectin-9, which has a high affinity for IgE in the blood. However, the amount of seaweed required to achieve this activity is quite high (12 g dry weight per person per day). Therefore, the purpose of this study was to search for food ingredients in vegetables that enhance type I hypersensitivity suppression effect when consumed together with fucoidan. As a result, the enhanced effect was observed in extracts from Welsh onions and onions among vegetables. When we compared the polyphenols in the vegetables that showed activity with those that did not, flavonols such as quercetin and kaempferol were found as candidates. When quercetin or kaempferol (100 μg each) were orally administered at the same time, even at amounts where fucoidan alone showed no anti-allergic activity, anti-allergic effects were observed. More interestingly, when both flavonols were combined and administered simultaneously at half the amount of each of the above flavonols (50 μg), while the fucoidan amount remained the same, a similar effect was observed as when each flavonol (100 μg) was administered alone. The simultaneous intake of fucoidan and vegetables containing high contents of quercetin or kaempferol may reduce fucoidan intake while maintaining the allergy suppression effect, suggesting the importance of food combination.

    Diets with high daily fat consumption are associated with excess weight. However, the effects of fat type and consumption timing on excess weight remain unclear. We investigated the selection of a 30% (w/w) fat diet of soybean oil (SOY), lard (LARD), and fish oil (FISH) on the metabolic parameters of mice. Male C57BL/6 mice were divided into the double SOY-box (w-SOY), SOY-box/LARD-box (SOY-vs-LARD), or SOY-box/FISH-box (SOY-vs-FISH) groups and allowed to selectively consume for 8 weeks. The total energy intake was similar for all groups, but the mice selectively chose to consume LARD over SOY and SOY over FISH. Body weight in the SOY-vs-LARD group was significantly higher than that in the w-SOY and SOY-vs-FISH groups. Additionally, minimal but selective consumption of an omega-3 fatty-acid-rich FISH diet at the end of the active period increased the physiological fatty acid compositions of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in the SOY-vs-FISH group; their metabolic parameters were also lower than the SOY-vs-LARD group. In conclusion, selectively consuming small amounts of fish oil at the end of the day may prevent excess weight compared with LARD consumption.

    The thinned immature fruit of the mango tree (Mangifera indica "Irwin") are regarded as waste products. In this study, we evaluated the effects of daily consumption of a hot-water extract of thinned immature mango fruits (TIMEx) on the dyslipidemia of apolipoprotein E-deficient (ApoE-/-) mice. ApoE-/- mice and wild-type BALB/c mice were fed a 20% fat diet containing 0%, 0.1%, or 1.0% TIMEx for 8 weeks. Their body mass, food intake, and water consumption were unaffected by the TIMEx. The 1.0% TIMEx supplementation significantly reduced serum triglyceride, but not total cholesterol concentration. This effect was significant in ApoE-/- mice, but less marked under normal conditions in wild-type mice. In addition, the circulating concentrations of three hormones that regulate metabolism, resistin, leptin, and glucose-dependent insulinotropic polypeptide, were reduced by TIMEx consumption, which may be involved in its effect to prevent hypertriglyceridemia. However, none of the concentrations of TIMEx reduced the size of atherosclerotic plaque lesions. In conclusion, daily consumption of TIMEx ameliorates hypertriglyceridemia but not hypercholesterolemia in genetically predisposed mice.

    In the present study, we evaluated the effects of daily consumption of raw (RR) or sun-dried (SDR) radishes (Raphanus sativus cv. YR-Hyuga-Risou) on apolipoprotein E-deficient (ApoE-/- ) mice. Daily consumption of RR for 16 weeks significantly decreased body weight gain in the both wild-type and ApoE-/- mice. The wild-type mice fed the SDR diet gained significantly less body weight than the ApoE-/- mice fed the same diet, although the ApoE-/- mice showed a trend toward decreased body weight gain. Consumption of both diets led to a marked decrease in visceral fat weight and serum triglyceride levels in ApoE-/- mice. Oral fat tolerance tests indicated that pretreatment with RR or SDR mitigated the increase in serum triglyceride levels seen after oil administration. In conclusion, we found that daily consumption of both RR- and SDR-containing diets can help us to prevent from dyslipidemia by inhibiting fat absorption.

    Quercetin has been shown to have anti-obesity effects, but it is unknown whether these effects can be transmitted from mothers to their progeny. In this study, we investigated whether maternal quercetin consumption during pregnancy has a protective effect on high-fat diet-induced hyper lipid levels and overweight in progeny. Female mice consumed a control diet or a diet containing 1.0% quercetin during breeding. The male progeny were then divided into four groups that were (1) sacrificed at postnatal day 3; (2) born to dams fed the control diet and also fed the control diet (C-C), (3) born to dams fed the control diet and then fed a 30% high-fat diet (C-HF), or (4) born to dams fed the Q-diet and then fed the HF diet (Q-HF). Maternal consumption of quercetin did not affect body weight or blood lipid parameters in either dams or neonates at postnatal day 3. After 13 weeks, the Q-HF group exhibited greater body and liver weights, and higher blood cholesterol levels than the C-HF group. However, the total cholesterol/ high density lipoprotein (HDL)-cholesterol ratios in the Q-HF and C-C groups remained similar. In conclusion, maternal quercetin consumption does not appear to protect the next generation from high-fat diet-induced hyper cholesterol level in the blood and liver, and consequently overweight, but may help regulate the total cholesterol/HDL-cholesterol ratio.

    Thinned immature fruit of the mango tree (Mangifera indica 'Irwin') are handled as waste. In this study, we conducted a 90-days toxicity study in male and female Sprague Dawley rats to evaluate the safety of a hot-water extract of thinned immature mango fruits (TIMEx) administered by oral gavage at doses of 500, 1000 and 2500 mg/kg body weight/day. Treatment did not result in death or changes in the behavior or external appearance of the animals. No alterations were observed in hematological or serum chemical parameters, urinalysis, food consumption, body weight gain or organ weights at the end of the treatment period, with the exception of higher mean corpuscular volume in male rats that received high doses and lower serum creatine phosphokinase levels in female rats that received medium doses. Under the conditions of this study and based on the toxicological endpoints evaluated, the no-observed-adverse-effect level (NOAEL) for TIMEx was 2500 mg/kg/day. The findings indicate that TIMEx is safe for consumption and should be investigated as a candidate food.

    The aims of this research communication were to investigate the in vivo tissue accumulation of phytanic acid (PA) and any changes in the tissue fatty acid profiles in mice. Previous in vitro studies have demonstrated that PA is a milk component with the potential to cause both beneficial effects on lipid and glucose metabolism and detrimental effects on neuronal cells. However, there is limited information about its in vivo actions. In this study, mice were fed diets containing either 0.00 or 0.05% 3RS, 7R, 11R-PA, which is the isomer found in milk and the human body. After 4 weeks, adipose tissue, liver and brain were harvested and their fatty acid profiles were determined by gas chromatographic analysis. The results showed that PA and its metabolite pristanic acid accumulated in the adipose tissue of PA-fed mice, and that dietary PA decreased the hepatic compositions of several saturated fatty acids such as palmitic acid while increasing the compositions of polyunsaturated fatty acids including linoleic acid and docosahexaenoic acid. However, dietary PA neither accumulated nor had a high impact on the fatty acid profile in the brain. These results suggested that dietary PA could exert its biological activities in adipose tissue and liver, although the brain is relatively less affected by dietary PA. These data provide a basis for understanding the in vivo physiological actions of PA.

    Background: Omega (ω) 3 fatty acid (FA) is a polyunsaturated FA (PUFA) that can modulate some mental statuses. However, most studies have not considered the functional differences between eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). We investigated the association among happiness, a sense of fulfillment and serum ω3 PUFA levels. Methods: Participants were 133 female staff from a hospital and nursing homes. Happiness was measured using the Japanese version of the subjective happiness scale (SHS); a sense of fulfillment was assessed using a visual analogue scale. Serum FA concentrations were measured. A partial correlation test and a regression model were applied. Results: The SHS scores showed significantly positive correlations with a sense of fulfillment, DHA% and EPA% (p < 0.05, p < 0.05 and p < 0.005, respectively), after controlling for age, BMI, menopause, snacking habits and leisure-time physical activities. A sense of fulfillment was significantly negatively correlated with α-linoleic acid%, and positively correlated with DHA% and EPA% (p < 0.05, p < 0.05 and p < 0.005, respectively), after controlling for the confounders. A regression model showed that a sense of fulfillment, EPA, and not stopping menstruation explained happiness (standardised beta, B = 0.18, p < 0.05; B = 0.24, p < 0.01; and B = 0.32, and p < 0.05, respectively), whereas age, BMI and snacking habits could not. Simultaneously, a regression model could not explain the association between DHA and happiness. Conclusion: Happiness was related with serum EPA%, a sense of fulfillment, and premenopause.

    Black soybean (Glycine max L.), a cultivar containing abundant polyphenols in its seed coat such as anthocyanins and flavan-3-ols, has been reported to possess various health benefits toward lifestyle diseases. In this review article, the safety evaluation of polyphenol-rich black soybean seed coat extract (BE) and absorption of BE polyphenols are summarized. Additionally, we describe the antioxidant activity of BE polyphenols and their ability to induce antioxidant enzymes. The health benefits of BE and its polyphenols, such as anti-obesity and anti-hyperglycemic activities through the activation of AMP-activated protein kinase and translocation of glucose transporter 4, respectively, are also discussed. Furthermore, we found that black soybean polyphenols were involved in the improvement of vascular function. These emerging data require further investigation in scientific studies and human trials to evaluate the prevention of lifestyle diseases using black soybean polyphenols.

    Polyphenols, a category of plant compounds that contain multiple phenol structural units, are widely distributed throughout the plant kingdom and have multiple benefits for human health including anti-obesity, anti-hyperglycemic, and anti-hyperlipidemic effects. Additionally, polyphenols have recently gained attention for their anti-stress effects. In this review article, we summarize physiological responses against exposure to stressors and discuss biomarkers for exposure to stressors that are widely used in animal studies and human trials. We also review commonly used animal models for evaluating anti-stress effects. Finally, we discuss recent findings related to the anti-stress effects of polyphenols evaluated in animal models and human trials, and their putative mechanisms. These emerging data require further investigation in scientific studies and human trials to evaluate the anti-stress effects of polyphenols and their potential use for the prevention of stress-related health problems.

    Background: There is growing interest in the possible role of circadian rhythms in feeding behavior and their effect on diet-induced obesity. However, it is unclear whether widely used purified diets are suitable for use as normal control diets for rodents undergoing time-restricted feeding studies. In the present study, we compared the effects of 4% and 10% fat diets on body mass gain and food consumption during time-restricted feeding (TRF).Methods: Ad libitum-fed male C57BL/6J mice had free access to AIN-93M (4% fat) or modified 10% fat diets, whereas TRF groups were only able to consume one of these diets twice daily, at ZT23:00–01:00 and ZT11:00–13:00, for 5 weeks.Results: Total energy consumptions by the TRF-4% and TRF-10% fat groups were 26.5% and 18.6%, respectively, less than that of the ad libitum-fed groups. The body mass gains by the 4% and 10% fat diet groups fed ad libitum were similar, but the gain by the TRF-4% fat group was markedly lower than that of the TRF-10% fat group. In addition, whereas the body mass gain by the TRF-10% fat group was similar to that of its ad libitum equivalent, the gain by the TRF-4% fat group was much lower than that of the equivalent ad libitum-fed group.Conclusion: We showed for the first time that a 10% fat diet (21.9% kcal from fat) is suitable for the maintenance of body mass gain during TRF of C57BL/6 mice.Keywords: ad libitum feeding, fat content, mouse, time-restricted feeding

    Salt intake is one of the most important environmental factors impacting hypertension onset. Meanwhile, the potential roles of the gut microbiome (GM) in altering the health status of hosts have drawn considerable attention. Here, we aimed to perform an observational study to investigate the impact of intestinal bacterial flora in hypertensive patients with low-salt or high-salt intake. A total of 239 participants were enrolled, and their gut microbiomes, clinical and demographic details, as well as physiological parameters pertaining to the renin-angiotensin-aldosterone system and inflammatory cytokine profiles, were examined. The participants were classified into four groups based on the presence of different enterotype bacteria, as determined via cluster analysis, and salt intake: low salt/GM enterotype 1, low salt/GM enterotype 2, high salt/GM enterotype 1, and high salt/GM enterotype 2. Results show that the prevalence of hypertension was significantly lower in the low-salt/GM enterotype 2 group (27%) compared to the low salt/GM enterotype 1 group (47%; p = 0.04). Alternatively, no significant differences were observed in hypertension prevalence between the two high-salt intake groups (GM enterotype 1 = 50%, GM enterotype 2 = 47%; p = 0.83). Furthermore, The low-salt/GM enterotype 2 was higher in the relative abundances of Blautia, Bifidobacterium, Escherichia-Shigella, Lachnoclostridium, and Clostridium sensu stricto than the low-salt/GM enterotype 1. differed significantly between the GM enterotypes. These results suggested that consumption of a low-salt diet was ineffective in regulating hypertension in individuals with a specific gut bacteria composition. Our findings support the restoration of GM homeostasis as a new strategy for controlling blood pressure and preventing the development of hypertension.

    Recent in vitro evidence suggests that the phytol-derived fatty acids, phytanic acid (PA) and pristanic acid (PrA), are components of animal products with the potential to cause both beneficial and harmful effects on human health. In this study, we investigated the in vivo tissue accumulation of PA and PrA and the changes in tissue lipid profiles, using mice fed a phytol-containing diet. After 4 weeks of treatment with a diet containing 1.0% phytol, plasma, adipose tissue, liver, and brain were collected and their lipid profiles were biochemically and gas-chromatographically determined. Dietary phytol caused PA and PrA accumulation in the adipose tissue and liver but not in the brain, and reduced plasma and liver triacylglycerol levels. Phytol intake also decreased the fatty acid concentrations in the adipose tissue, especially polyunsaturated fatty acids such as linoleic acid, but increased the concentrations of these fatty acids in the liver. However, dietary phytol had a low impact on the brain lipid profile. This study suggests that dietary phytol intake caused accumulation of PA and PrA and modified lipid profiles in the adipose tissue and liver, but that the brain is an insusceptible tissue to dietary phytol-induced changes.

    BACKGROUND: Chronic low-grade inflammation and oxidative stress are commonly observed in persons with depression or depressive symptoms. We explored the degree of depressive symptoms under psychological stress in relation to serum LDL oxidation, inflammatory markers, and fatty acid (FA) distribution among female population. The purpose of this study was to identify peripheral factors that are related to depressive symptoms, and to assess how each factor is related to depressive symptoms. METHODS: 133 female workers in a hospital and nursing homes were recruited in Japan. Depressive symptoms were assessed using the Japanese version of the Centre for Epidemiologic Studies Depression Scale (CES-D), and perceived stress was assessed using the visual analogue scale. Cytokine levels and oxidation rate of LDL cholesterol (ox-LDL/LDL) were measured as indices of inflammation and oxidation. Omega-3 FA distribution was also measured. Path analysis and hierarchical regression analysis were used to determine if each factor was predictive of depressive symptoms. RESULTS: It was identified that serum ox-LDL/LDL was positively connected with depressive symptoms, but was more strongly related to perceived psychological stress. Elevated serum IL-6 was positively correlated with depressive symptoms, though the effect was partly transmitted via ox-LDL/LDL. Additionally, serum ω3 PUFAs were inversely associated with depressive symptoms independently of IL-6 or ox-LDL/LDL. CONCLUSION: Although this study is unlikely to fully explain the causes of depressive symptoms, it suggests that psychological stress and somatic factors such as inflammation, oxidation and nutrition are related to depressive symptoms. These findings suggest the therapeutic potential of lifestyle targets to alleviate the identified depression risk factors, anti-oxidative therapies, anti-inflammatory therapies and nutritional interventions to prevent depression.

    Previously, we reported that the frequency of micronucleated reticulocytes (MNRETs) in the peripheral blood of male C3H/He mice intraperitoneally administered ethylnitrosourea (ENU) (25 mg/kg body weight) in the dark period (zeitgeber time, ZT15) was higher than in the light period (ZT3). In this study, to clarify the mechanism underlying this phenomenon, we investigated the differences in micronucleus (MN) induction observed between ZT3 and ZT15 using five chemicals, methylnitrosourea (MNU), ethylmethane sulfonate (EMS), mitomycin C, cyclophosphamide and vincristin. MNU and EMS, monofunctional alkylating agents, showed higher frequencies of MNRETs in the ZT15 than the ZT3 treatment similar to ENU. However, no differences were observed for the other chemicals. In the comet assay, more DNA damage was induced by ENU in the ZT15 than the ZT3 treatment. Furthermore, the plasma erythropoietin (EPO) level, a known effector of MN induction with anti-apoptotic activity mediated by Bcl-xL expression, was higher in the dark than in the light period. EPO did not increase the frequency of MNRETs. However, in the ENU treatment group at ZT3 following EPO injection a significant increase of MNRETs was observed similar to the ZT15 treatment. Higher expression of apoptosis-related genes such as Bcl-xL was induced in bone marrow cells from mice treated with ENU at ZT15 compared with ZT3. From these results, it was speculated that the differences in MN induction in the peripheral blood of mice exposed to monofunctional alkylating agents such as ENU depend on apoptotic or anti-apoptotic conditions related to the circadian rhythms of EPO in bone marrow.

    Blueberry leaf may contain multiple compounds with beneficial effects. We conducted a 90-day toxicity study in rats to evaluate the safety of consuming the leaves of rabbiteye blueberry (Vaccinium virgatum Aiton; RB species). Powdered leaves were administered daily by oral gavage at doses of 500, 1000, and 2500 mg/kg body weight to male and female Sprague-Dawley rats for 90 days. Treatment did not result in death or changes in the behavior and external appearance of the animals. No alterations were observed in hematological and serum chemical parameters, urinalysis, food consumption, body weight gain, or absolute and relative organ weights at the end of the treatment period, with the exception of some leukocyte percentages in male rats treated with 500 and 1000 mg/kg blueberry leaf powder. The findings indicate that rabbiteye blueberry leaf is safe for consumption and should be investigated as a candidate functional food.

    Recent studies indicate that patients with depression have increased concentrations of serum pro-inflammatory cytokines. However, studies of IL-17 and studies on community-dwellers are few. The purpose of this study was to investigate serum cytokine levels, especially IL-17A, among subjects with high and low depressive symptoms of a general population. The participants comprised 20 female community-dwellers aged 40 years or older who contributed to a Shika study in Ishikawa, Japan. Ten participants who showed higher and ten who showed lower depressive symptoms among 208 females assessed by the Japanese version of the Centre for Epidemiologic Studies Depression Scale (CES-D) were selected for this study. Serum samples were analyzed for TNF-alpha, IL-6, IL-10, IL-12, and IL-17A using a multiplex Luminex analysis. For the comparison between the high and low depressive groups statistically, linear regression analyses were applied. The serum level of IL-17A was significantly higher among the high depressive participants (p < 0.05) even after controlling possible confounders, whereas there were no differences in TNF-alpha, IL-6, IL-10, or IL-12 between the high and low depressive groups. Our findings supported an association between serum IL-17A levels and depressive symptoms. Peripheral IL-17A immune response may be a preventive and treatment target for depression.

    Flavonoids are a major group of natural products that are widely distributed in dietary plants, exerting beneficial functions such as antioxidant activity. The purpose of this study was to propose protocols for a flavonoid-poor menu satisfying the daily nutrient requirements for use in clinical studies to evaluate the bioavailability and functional activities of target flavonoids. First, a ranking of flavonoid-rich foodstuffs consumed in Japan was compiled based on their standard daily consumption, and it was decided not to use foodstuffs containing more than 0.1 μmol flavonoids per dish for preparation of meals. Two daily menus were then developed using other foodstuffs satisfying the dietary reference intakes for Japanese males. Furthermore, we confirmed that the target flavonoid could not be detected (<0.2 μmol per meal) in the developed menus, indicating that volunteers would be able to achieve the Dietary Reference Intakes for Japanese, 2005, with negligible flavonoid intake by consuming the meals we had developed. In conclusion, we have been able to propose protocols for preparation of flavonoid-poor meals that would be useful for clinical studies.

    Precursors of nutraceuticals are synthesized through metabolic pathways common to higher plant species. Although light-emitting diode (LED) has recently been applied to "plant factories", we know little what enzyme genes or substances in metabolic pathways are enhanced in response to LED irradiance.
    Arabidopsis thaliana and other Cruciferae plants were exposed to blue LED (470 nm), red LED (660 nm), blue LED + red LED, or blue LED (pulsed) at 110-240 μmol m-2 s-1, and subjected to DNA array analysis (Affymetrix GeneChip Arabidopsis ATH1) or metabolomics (Agilent 6510 Accurate-Mass Q-TOF LC-MS/MS and others). Expression of gene for biosynthesis of lignans and flavonoids have been enhanced by blue LED, leading to enrichment with kaempferol glycosides and anthocyanins. Gene for chalcone synthase (AtCHS) has been ectopically expressed and resulted in accumulation of kaempferol glycosides.

    Journal of Toxicological Sciences, Jun. 2016, Japanese, 【背景】近年、光環境の変化が生体に及ぼす影響について様々な研究が行われている。我々は以前、アルキル化剤であるエチルニトロソウレア(ENU)の投与時刻について検討し、明期投与より暗期投与でのマウス末梢血中の小核誘発率が高いことを報告した( J. Toxicol. Sci., 37, 427-430, 2012)。一方、暗期に1時間光を曝露することにより、マウス肝臓のPai-1遺伝子発現が上昇することを我々は報告している( Exp. Anim. 63(3), 331-8, 2014)。そこで、本研究では、①DNA損傷を誘発する他の化学物質の明期投与および暗期投与が小核誘発に及ぼす影響と②暗期の光曝露が肝臓のPai-1遺伝子発現に及ぼす影響に注目し、動物飼育における光環境の影響についてC3H/He Slcを用いて検討した。【方法と結果】①非アルキル化剤と、ENU以外のアルキル化剤を明期と暗期にそれぞれ投与し、小核誘発率について検討した。その結果、アルキル化剤投与群では、ENUと同様に明期投与よりも暗期投与での小核誘発率が高値を示し、非アルキル化剤投与群ではそのような違いはみられなかった。その違いは化学物質が誘発するDNA損傷や修復のタイプに起因することが推察された。②暗期の光曝露(2時間)により、血中アドレナリン濃度とコルチコステロン濃度が上昇し、肝臓・心臓におけるPai-1遺伝子発現と血中PAI-1の上昇が見られた。また、アドレナリンβ2受容体遮断薬により肝臓における遺伝子発現変化は抑制された。よって、光曝露により交感神経系/SAM系/HPA系が活性化しPAI-1の上昇を引き起こす可能性が考えられた。【まとめ】以上より、光曝露により化学物質に対する感受性が異なったり、ストレス応答が誘導され得ることがわかった。よって、動物飼育において、光環境の影響に十分注意する必要がある。

    J Toxicol Sci, Jun. 2013, Japanese, ヒトを含めたほとんどの哺乳動物の行動や生理的な変化は,約24時間を周期とした概日リズムを持っている。近年,この概日リズムに関連した生体反応の違いについて,時間薬理学や時間栄養学などの様々な分野で研究が行われ,臨床においては抗がん剤の時間治療も実施されている。安全性試験の分野では,ヒトでの安全性を評価するためにげっ歯類を用いた試験で得られた結果をヒトに外挿しているが,一般的な実験では,被験物質の投与や各種検査をマウスやラットなど夜行性動物の睡眠期にあたる明期に実施することが多い。一方,ヒトでは日中の活動期に種々の化学物質に曝露されることが多い。
    我々は,安全性試験における投与時刻の影響を検討する目的で,遺伝毒性試験の一つである小核試験を行った。明期(睡眠期)または暗期(活動期)に,変異原性を有するアルキル化剤であるN-エチル-N-ニトロソウレア(ENU)を8~9週齢の雄性C3Hマウスに腹腔内投与し,その後経時的に尾静脈から採血して末梢血中の小核誘発頻度を調べた。その結果,暗期の投与では明期の投与よりも小核誘発頻度が有意な高値を示した。この機序を解明するために,骨髄細胞におけるp53標的遺伝子(p21,cyclin G1,bax)の発現をリアルタイムPCRにより検討した。また,フローサイトメーターを用いて細胞周期(G0/G1,S,G2期)を測定した。その結果,明期,暗期のいずれの投与時期においてもp53標的遺伝子の発現がENU処置により誘導されることが確認され,cyclin G1およびbaxについては,明期の投与で暗期の投与よりも発現量が高値傾向を示した。細胞周期については明らかな変化は認められなかったが,現在,細胞周期に対する影響に関して詳細な検討を進めており,これらの結果についても報告する。

    Purple yam (Dioscorea alata L.), which are distributed in tropical and subtropical regions, is characterized by color and viscosity. Previous studies have shown that acylated anthocyanins are presented in purple yam and the components have stronger antioxidant activity than that of non-acylated anthocyanins. We isolated four new acylated anthocyanins (Compounds 1~4) and four known anthocyanins (Compounds 5~8) from the purple yam collected in Philippine by various column chromatographic techniques. By using MS and NMR spectroscopic techniques, three new compounds (1, 3, 4) were determined to be cyanidin type and the other (2) was peonidin type anthocyanins. Antioxidant activity of the purple yam extracts and isolated anthocyanins were tested by FRAP and ORACFLassays. Antioxidant activity of the purple yam extracts showed stronger than that of the purple sweet potato extracts in FRAP assay. Compound 6 with two sinapic acids in the structure had the most potent antioxidant activity in isolated anthocyanins.

    J Toxicol Sci, Jun. 2011, Japanese, 【緒言】アラニンアミノトランスフェラーセ゛(ALT)は肝障害マーカー酵素であるが,肝障害を伴わずに変動することが知られている。我々は前回大会で,制限給餌で飼育したラットにおける血中ALT活性の変動には,肝臓及び小腸中ALT活性の寄与が大きいこと及び制限給餌により肝臓中ALT活性が上昇することを報告した。本発表では,制限給餌で飼育したラットにおける肝臓中ALT活性の上昇機序について報告する。【方法】雄性SDラットを自由摂取群(ALF群)あるいは制限給餌群(SF群,1日4時間,9:00-13:00に給餌)の2群に分け35日間飼育した。制限給餌開始前日の13時から絶食を行い,制限給餌開始日をD1としてD1,3,7,14及び35に血液を採取し血中ク゛ルコース及びアラニン濃度を測定した。また,D1及びD35に採取した肝臓を用いて,アミノ酸,糖,脂肪酸酸化に関わる酵素の遺伝子発現量をRT-PCR法で測定した。【結果及び考察】SF群の血漿中ク゛ルコース濃度は,D1,3,7及び14でALF群と比較して低値を示したが,Day 35の血漿中ク゛ルコース濃度は,ALF群との間に差は認められなかった。SF群の血漿中アラニン濃度は,D1,7,14及び35でALF群と比較して低値を示した。SF群の肝臓における脂肪酸及び糖新生に関わる酵素の遺伝子発現は,D1及びD35ともにALF群に比べて増加した。一方,解糖系酵素の遺伝子発現はD1で抑制がみられたが,D35ではALF群と比較して差はみられなかった。以上の結果から,SFラットでは肝臓での脂肪酸酸化及び糖新生が亢進し,アラニン及びク゛ルコースの血中濃度が変動したと考えられた。ALTが糖新生に関わる酵素であることを考慮すると,肝臓における脂肪酸酸化及び糖新生の亢進に伴い肝臓中ALTの生合成が亢進したことにより肝臓中ALT活性が上昇したと考えられた。

  • ストレス応答が過敏になる夜間の生体応答制御を企図した食事の効果検証

    ストレス社会の蔓延により,精神障害を抱える日本人の絶対数が増加している。そのため,精神障害予防を目指した新たな制圧戦略が必要である。本研究は,ファイトケミカルの抗ストレス効果を,ストレスに対する生体応答機構が刻む日内リズムとファイトケミカルの摂取時刻の観点から検証し,最終的に抗ストレス効果を有するファイトケミカルを含んだ食事をいつ摂取すべきかの提案を目指すものであり,令和3年度は以下の成果を得た。 1)マウスを用いた試験系により,ストレスに対する生体応答機構に対する生命の時間軸の影響を調べた結果,加齢に伴いストレス応答性が上昇するプロテインXを見出した。 2)栄養成分の代謝吸収に関与する因子の肝臓中遺伝子発現の日内リズムを評価した結果,特に明確なリズムを刻んだ脂質代謝系に着目することにした。次に,異なる脂肪酸組成の油を異なる時間帯に経口投与したところ,その体内動態が投与時刻により異なることを確認した。さらにその作用は,単回投与よりも日常的に摂取することでより明確になることを見出した。 3)代表的なフラボノイドの一つであるケルセチンを被験ファイトケミカルとして用い,異なる配糖体を経口投与した時に,胃―回腸における通過速度に違いあることを見出した。 4)腸内細菌叢解析を開始した。

  • 脂質摂取がアルツハイマー型認知症状に与える影響と食品によるその制圧

    超高齢社会を迎えている現代社会において, アルツハイマー型認知症(以下, ADと略)の予防・治療法を確立することは, 喫緊に取り組むべき課題の一つである。近年の研究において, 脂質の過剰摂取がADの危険因子であること, 血中コレステロール高値がADの発症と相関することが報告なされたことから, 脂質代謝系を調節できる食事因子を日常的に摂取し続けることで, ADの発症を予防できる可能性がある。本研究では, ADモデルであるB6SJL-Tgマウスを用い, 脂質摂取がADの病態に与える影響を, βアミロイドタンパク質の脳への蓄積, 学習・記憶・不安などの行動変化を主軸とした脳機能, 血・脳・臓器中脂質量等の変化を時間軸で明らかにすることを目的として推進し, 令和元年度は以下の成果を得た。 1)野生型の雄性B6マウスと雌性SJLマウスを交配し, 次世代雌性マウスを得た。 2)雄性B6SJL-Tgマウスと1で作製した雌性マウスを交配し, F1マウスを得た。 3)さらに交配を継続し, F2マウスを本試験に供した。本試験では, コントロール食(4%脂質)あるいは高脂肪食(24%)を4ヶ月間自由摂食させた。

  • Adverse effect on age-related brain function by environmental and social stress and a preventive role of food factors

    Effects of aging on social stress, such as isolation and confrontation, induced physiological responses were investigated using young, middle-aged, and old mice (C57BL/6N and BALB/c). A stress response via activated HPA axis was induced in each aged mice. It was suggested that isolation stress for a long period may affect the lipid metabolism and induce intracerebral inflammation, especially in middle aged mice. It was found that the expressions of two genes in the liver or the hippocampus were reduced or induced by social stress. These two genes could be used as a new stress related marker.

  • Investigation of polyphenol paradox

    In the studies for the disease prevention effect of polyphenols, we propose a new terminology, the "polyphenol paradox", that the results observed in cultured cell and animal studies are not observed in the clinical trials. To clarify this paradox, we performed 1) pharmacokinetic analysis considering biological rhythm: 2) elucidation of target molecule in the body: 3) elucidation of function considering tissue-tissue crosstalk: 4) elucidation of function in the nanomolar concentration range. We have obtained results on certain polyphenols and their metabolites at the low-concentration that can exist in the body to exert various bioregulatory functions: the molecular mechanism of these functions, tissue-tissue tissue crosstalk, target molecules, optimal ingestion timing, and stress response. We have built a research base foundation for developing the obtained results and applying them to the future clinical trials.

  • Mother-infant interaction of functional flavonoids

    In this project, we found that daily consumption of flavonoids increases their absorption rate, and also one of major functions is the effect of lipid metabolism. Therefore, we employed quercetin as candidate flavonoids, and evaluated it’s mother-infant interaction. Consequently, we obtained the following aspects: 1) hepatic triglyceride levels were significantly decreased in the infants from the mother consuming quercetin during the pregnancy, and this effect was remarkable in the groups consuming high-fat diet; 2) mice from mother consumed quercetin were sensitive for high-fat induced overweight comparing with mice from mother not consumed quercetin. In conclusion, our results clearly indicate the importance for mother-infant interaction of functions existed in dietary flavonoids.

  • Inflammation and oxidation in depressive states

    Low-grade inflammation and oxidative stress are observed in persons with depressive symptoms (Dep). We explored Dep under psychological stress in relation to serum LDL oxidation rate (ox-LDL/LDL), inflammatory markers, and fatty acids (FAs) among female populations. We aimed to identify peripheral factors that are related to Dep. Serum ox-LDL/LDL was positively connected with Dep, but was more strongly related to perceived psychological stress. Serum IL-6 was positively concerned with Dep, though the effect was partly intervened by ox-LDL/LDL. Further, serum ω3 FAs were inversely associated with Dep independently of IL-6 or ox-LDL/LDL. In addition, serum IL-17A was higher among high depressive participants. These findings suggest the therapeutic potential of lifestyle targets to alleviate the identified depression risk factors, anti-oxidative therapies, anti-inflammatory therapies and nutritional interventions to prevent depression.

  • Discovery of novel sulfated bioregulatory molecules acting on neural tissue receptors

    In this study, we focused on the functional control of bioregulatory molecules by sulfation as a metabolic mechanism. First, enzymatic preparation method of sulfated bioregulatory molecules were developed. Furthermore, we conducted research aimed at developing analysis technology for cell surface receptor-mediated signal transduction and elucidating its physiological function in cultured cells. Specifically, using E. coli that expressing human cytosolic sulfotransferase, we have established a biotechnology method to prepare a sulfated metabolite of a bioregulatory molecules such as food components. The obtained sulfated metabolite confirmed the chemical structure by NMR. Furthermore, a luciferase assay to evaluate GPR30 receptor-mediated activity was established, and the sulfated metabolites were evaluated for functional analysis.

  • Relationship between biological functions and sleep-movements exerted in leguminous plant

    Soy bean, one of the major leguminous plants, are widely recognized to exert sleep-movement rhythm. In this study, we evaluated the relationship between biological functions and its sleep-movements, and obtained the following results. (1) soy bean leaf indicated clear sleep-movement rhythm, (2) antioxidant activity was stimulated the beginning of dark period, (3) gene expression of LUX indicated typical diurnal rhythm, (4) isoflvone amounts decreased at ZT6, and (5) gene expression of C4H concerning isoflavone synthesis reached at maximum at ZT6 in a legume.

  • Database about circadian system regulating bioavailability of food factors, and their applications.

    Circadian profiles, for example typical clock genes such as Bmal1, Clock and Cry, and transporter genes such as Sglt1, Pept1 and Mdr1, existing in the liver and small intestine were analyzed in mice. Following, database about circadian system regulating bioavailability of food factors were summarized. These circadian system including clock genes was affected by the environmental factors, for example nocturnal light and isolation stress exposure into mice. Furthermore, the effects of consumption timing at active- or inactive-phase on bioavailability of bilberry anthocyanins were evaluated. Their gastric emptying rate was significantly faster upon administration at the active-phase than that of the inactive-phase. Their amounts appeared in the plasma suggested also different time-dependent property, implying that the intensity of beneficial effects of anthocyanins might be different in the alimentary canal and/or physiological environment according to their consumption timing.

  • Can beneficial effects of food factors reach embryo via mother's body?

    We evaluated whether beneficial effects existed in the foods and food factors can reach embryo and infant via mother's body, and obtained the results indicating that some factors, such as bilberry anthocyanins, might affect on the lipid metabolic pathway, but not the antioxidative system, in the liver on embryo, when mothers were consuming food factors. On the other hand, extracts of bitter cucumber, which has been reported to reduce the hepatic triacylglycerol level in the adult animals, did not exert any effects on hepatic lipid metabolism in the mice embryo. We are continuing the studies to obtain further evidences on this research topics.

  • Environmental pollutions and ecological effects of novel halogenated aromatic hydrocarbons

    In the present study, we focused the halogenated polycyclic aromatic hydrocarbons (halogenated PAHs) that is one of non-regulated pollutants, and investigated the occurrences in the environment and the biological effects. The surveys of chlorinated PAHs (ClPAHs) ambient air in 14 sites in Japan, 1 site in Korea and 1 site in China showed that ClPAHs is universally present in the air since some of that were detected from all air samples. In addition, the surveys of ClPAHs in Asian sediments also showed the universalistic pollution. The biological effects were investigated by oral administration of ClPAH in rat, resulting that chlorination of PAH could enhance significantly induction of specific drug-metabolizing enzyme.

  • Effect of light and dark dosing time on biological response to environmental chemicals in mice

    Any differences in the frequencies of micronucleus induction in peripheral blood reticulocytes(MNRETs) of mice exposed to BaP between light and dark dosing times were not observed. However, the expression of lipid metabolism related genes such as Fasn in liver was dramatically increased in treatment group at ZT15(darkphase). On the other hand, MNRETs of mice treated with ENU at ZT15 were significantly higher than those in mice treated at ZT3 (light phase). The expression of p53-responsible genes such as Cyclin G1 in the bone marrow cells was higher in treatment group at ZT3 than that at ZT15. DNA repair might be promoted at light phase. These results suggest that toxic sensitivity to chemicals is different between the light and dark phase.

  • Investigation of common markers to depressive symptoms and metabolic syndrome-for the prevention and intervention studies

    To investigate the common indicators to metabolic syndrome and depressive symptoms, a cross-sectional study was carried out on women and men in general population. Different serum indicators concerning lipid-or glyco-metabolisms were related to depressive symptoms between male and female population. In addition, an antioxidant appeared to reduce the depressive symptoms. Genotypes in the fatty acid synthase were found to be associated with depressive symptoms. Some common indices to metabolic syndrome and depressive symptoms were presented.

  • Investigation of pre-biomarker for social stress-inducible obesity in mice

    Mice were singly housed with reduced bed-volume for 3 months. Their body and visceral fat weights were significantly increased in combination with hepatic hypertrophy because of lipid deposition comparing to the control (5 mice/cage). The alterations of blood creatine kinase and plasminogen activator inhibitor type 1 (PAI-1) levels were suggested to might be used as pre-biomarker for social stress-inducible obesity. Additionally, PAI-1 increased under stressful events might not affect on sleep disturbance.

  • Combined effects of environmental chemical stress and social stress oninitiation of carcinogenesis

    単独隔離の飼育条件によりマウス(BALB/c、5週齢)にマイルドなストレスを30日間負荷し続けると、肝臓中の脂肪酸合成に関する遺伝子の発現量が上昇、脂肪酸分解に関する遺伝子の発現量が減少し、その結果、脂肪蓄積を伴った肝肥大が誘発されることを明らかにした。一方、BaPを投与すると、ストレス負荷群では、肝臓中のDNA付加体が減少しにくいことがわかった。これらの結果より、社会的ストレス負荷時にBaPのような脂溶性の高い発がん物質が体内に取り込まれると、肝臓に蓄積して発がんのイニシエーション過程を促進する可能性が示唆された。
