- Scope Malaria remains one of the most important infectious diseases in the world. Allyl isothiocyanate (AITC) is a main ingredient of traditional spice Wasabia japonica, which is reported to have anti‐bacterial and antiparasitic activities. However, there is no information on effects of AITC against malaria. The present study investigates the anti‐malarial activity of dietary AITC in vivo and that of AITC metabolites in vitro. Methods and results The ad libitum administration of 35, 175, or 350 µM AITC‐containing drinking water to ICR mice significantly inhibit the parasitemia induced after infection with Plasmodium berghei. On the other hand, after single oral administration of AITC (20 mg kg−1 body weight), N‐acetyl‐S‐(N‐allylthiocarbamoyl)‐l‐cysteine (NAC‐AITC) as one of the AITC metabolites displays a serum Cmax of 11.4 µM at a Tmax of 0.5 h, but AITC is not detected at any time point. Moreover, NAC‐AITC shows anti‐malarial activity against Plasmodium falciparum in vitro, and its 50% inhibitory concentration (IC50) against parasitemia is 12.6 µM. Conclusions These results indicate that orally administered AITC is metabolized to NAC‐AITC and exerts anti‐malarial activity against malaria parasites in blood, suggesting that the consumption of AITC‐containing food stuffs such as cruciferous plants may prevent malaria.Wiley, Sep. 2023, Molecular Nutrition & Food Research, 67(21) (21), English[Refereed]Scientific journal

- Boysenberry (Rubus loganbaccus × baileyanus Britt.) is one of the popular berries in Western countries. In the present study, the effect of the boysenberry juice on ferric nitrilotriacetate (Fe-NTA)-induced renal injury was investigated. ICR mice (7 weeks old, male) were ad libitum administered boysenberry juice for 1 week, and were intraperitoneally injected Fe-NTA (5 mg Fe/kg body weight) as an oxidant to renal. The consumption of boysenberry juice suppressed the Fe-NTA-increased thiobarbituric acid reactive substances (TBARS) in kidney and blood urea nitrogen (BUN). - Objective: The effects of 6-MSITC on cell cycle progression were investigated in quiescent mouse epidermal JB6 cells. Background: 6-Methylsulfinylhexyl isothiocyanate (6-MSITC) derived from wasabi (Wasabia japonica) has been reported to prevent tumor development in vivo. Material and methods: Treatment with epidermal growth factor (EGF) to quiescent JB6 cells, which were serum-starved for 36 h, promoted cell cycle progression from the G0/G1 phase to the S phase. Effects of pretreatment with 6-MSITC on cell cycle progression were estimated by flowcytometry and real-time RT-PCR. Results: Pretreatment with 6-MSITC at 0.25-1.0 μg/ml prior to the growth stimulation with EGF significantly inhibited cell cycle progression. Pretreatment with 6-MSITC inhibited the gene expression of DNA synthesis-related proteins cyclin A2, dumbbell former 4, and proliferating cell nuclear antigen. Conclusion: These results showed that 6-MSITC inhibits cell cycle progression in quiescent cells, accompanied by the inhibition of gene expression of DNA synthesis proteins.Sciencedomain International, Jun. 2021, Annual Research & Review in Biology, 36(6) (6), 19 - 28, English[Refereed]Scientific journal
- Artepillin C, 3, 5-diprenyl-4-hydroxycinnamic acid, is one of the bioactive constituents in Brazilian propolis. In the present study, the anticarcinogenic activity of this compound was investigated in human hepatoma HepG2 cells. Artepillin C inhibited the cell proliferation in a dose- and time-dependent manner accompanied by G0/G1 phase arrest in the cell cycle. This compound caused a decrease in the phosphorylation levels of the retinoblastoma protein at Ser780 and Ser807/811 and a decrease in the kinase activity of the cyclinD and CDK4 complex without any change in these protein levels. Artepillin C increased the protein level of p27Kip1, known as a CDK inhibitor. This up-regulation was regulated by both the transcriptional and post-transcriptional levels, i.e., the treatment increased the mRNA of p27Kip1 and decreased the proteosome activity. Thus, artepillin C induces cell cycle arrest at G0/G1 phase accompanied by up-regulation of p27Kip1, resulting in the inhibition of cell proliferation in HepG2 cells. - Laminaria japonica is traditionally eaten in Japan as a beneficial food for thrombosis. The alga contains two specific ingredients, a xanthophyll fucoxanthin (FX) and a polysaccharide, F-fucoidan (FD). The aim of the present study was to investigate whether FX or FD exhibited anti-thrombotic effects. For this purpose, three types of capsules, containing 1 mg FX, 400 mg fucoidan, and both, were prepared from the alga and administered to volunteers for 5 weeks. The dose of FD or FD+FX significantly shortened lysis time (LT) of the thrombus measured by a global thrombosis test in the blood, but FX did not. Examining the mechanism, dietary FD increased H2O2 and the secretion of prostacyclin (PGI2), a potent inhibitor of platelet aggregation, in the blood, although FD was under the detection limit in the blood, determining with its monoclonal antibody. Furthermore, in mouse experiments, dietary FD was totally excreted into the faeces and was not incorporated into the blood. We then employed a co-culture system of a Caco-2 cell monolayer with fresh human blood. The addition of FD to Caco-2 cells stimulated the expression of NADPH oxidase 1 (NOX1) and dual oxidase 2 (DUOX2) mRNA and secreted H2O2 onto the blood side accompanied by a significant increase in serum PGI2 production. These effects were invalidated by the combined addition of FD with its monoclonal antibody. The results suggested that dietary FD stimulated the expression of H2O2-producing enzymes in intestinal epithelial cells and released H2O2 into the blood, which played a signalling role to increase PGI2 production and then shortened LT for thrombi.Sep. 2013, The British journal of nutrition, 110(5) (5), 880 - 90, English, International magazine[Refereed]Scientific journal

- Dietary fucoxanthin has been reported to exert several physiological functions, and fucoxanthinol is considered to be the primary active metabolite of fucoxanthin. However, there is no information about the pharmacokinetics of fucoxanthinol in human subjects. In the present study, eighteen human volunteers were orally administered kombu extract containing 31 mg fucoxanthin, and their peripheral blood was collected 5 min before and 0·5, 1, 2, 4, 8 and 24 h after the treatment. Plasma fucoxanthinol concentrations were measured by HPLC, and the pharmacokinetics of fucoxanthinol were as follows: maximum concentration, 44·2 nmol/l; time at maximum concentration, 4 h; terminal half-time, 7·0 h; area under the curve (AUC) for 1-24 h, 578·7 nmol/l × h; AUC(∞), 663·7 nmol/l × h. In addition to fucoxanthinol, we also attempted to detect amarouciaxanthin A, a hepatic metabolite of fucoxanthinol, using HPLC, but it was not present in the volunteers' plasma. On the other hand, a peak that was suspected to represent the cis-isomer of fucoxanthinol was found in the HPLC chromatogram. - To clarify the mechanism of the antiallergic activity of Agaricus blazei Murill extract (ABME), the present paper used an in vivo allergy model and an in vitro intestinal gut model. During OVA sensitization, the serum IgE levels decreased significantly in ABME group. Interleukin (IL)-4 and -5 produced from OVA-restimulated splenocytes was significantly decreased, and anti-CD3ε/CD28 antibody treatment also reduced IL-10, -4, and -5 production and increased IFN-γ production in ABME group. These results suggest that oral administration of ABME improves Th1/Th2 balance. Moreover, a coculture system constructed of Caco-2 cells and splenocytes from OT-II mice or RAW 264.7 cells indicated that the significant increases in IFN-γ production by ABME treatment. Therefore, it was concluded that the antiallergic activity of ABME was due to the activation of macrophages by epithelial cells and the promotion of the differentiation of naïve T cells into Th1 cells in the immune.2011, Evidence-based complementary and alternative medicine : eCAM, 2011(. pii) (. pii), 532180, English, International magazine[Refereed]Scientific journal

- The synergistic effect of food additives or food colors on the toxicity of 3-amino-l, 4-dimethyl-5H-pyrido[4, 3-b]indole (Trp-P-1) was investigated using primary cul-tured rat hepatocytes. When hepatocytes from rats fed a standard diet were treated with a mixture of four major food additives (sorbitol, sodium L(+)-glutamate, benzoic acid, and propylene glycol) or a mixture of six typical artificial food colors (erythrosine, allura red, new coccine, brilliant blue, tertrazine, and fast green), the in vitro treated food-color mix-ture itself showed cytotoxicity; the reduction of cell viability and decreases in the activities of gluconeogenesis and ureogenesis. The food-color mixture enhanced cytotoxicity of Trp-P-1 obviously. We then investigated the effects of in vivo-dosed food additives or food colors on Trp-P-l-caused toxicity. Hepatocytes were isolated and cultured from rats fed a diet contain-ing a mixture of food additives or a mixture of food colors with half the amount of their re-spective acceptable daily intake for 4 wk. Trp-P-1 was administered to the hepatocytes at various concentrations for 12 h. Synergistic effects of in vivo-dosed food additives and food colors were not observed on Trp-P-l-caused cytotoxicity as estimated by a loss of cell viabil-ity and the reductions of DNA and protein syntheses. On the contrary, we have observed that in vivo administered food colors synergistically facilitated to reduce the activities of glu-coneogenesis and ureogenesis in Trp-P-l-treated hepatocytes. These results suggest that the daily intake of artificial food colors may impair hepatic functions such as gluconeogenesis and ureogenesis, when dietary carcinogens are exposed to the liver cells.Center for Academic Publications Japan, Jun. 2000, Journal of Nutritional Science and Vitaminology, 46(3) (3), 130 - 136, English

- Use of Pulverized Cold-pressed Flaxseed Meal as a Food Ingredient and Antioxidative Potency of Flaxseed LignansFlaxseed has been cultivated for at least 8,000 years in Europe and Asia, mainly for use in industrial paint oil and polymers, and partly for cereals and baked goods. The consumption of flaxseed as a food has recently in-creased dramatically, as it contains a large amount of the n-3 fatty acid, α-linolenic acid, dietary fiber, and flaxseed lignan. We report a new method to improve the processing suitability of flaxseed as a food ingredient. An impact mill with classifier was used to grind cold-pressed flaxseed meal into a fine powder which was stable against oxidation. This finely ground cold-pressed flaxseed meal was suitable for making baked goods such as cookies and bread. Flaxseed lignan in the cold-pressed meal was stable under heat treatment, and the lignan glucoside in flax-seed and its derivatives showed strong antioxidative potency.日本調理科学会, 2007, Journal of cookery science of Japan, 40(2) (2), 74 - 81, Japanese

- 日本応用糖質科学会平成27年度大会, Sep. 2015, Japanese, 奈良, 【目的】これまでに我々はマコンブに含まれるF-フコイダンがⅠ型アレルギー抑制効果を有しており、その効果にガレクチン9が関与していることを明らかにしてきた。ガレクチン9はβガラクトシドを認識するレクチンの一種で、近年IgEと結合することでIgEの肥満細胞への結合を阻害することが報告されている。今回、我々は実際にF-フコイダンによりガレクチン9が血中で増加してⅠ型アレルギー症状を抑制しているのかを検討した。 【方法】F-フコイダン(200 μg/日)を4日間胃内強制投与した。抗原特異的IgEを尾静脈から投与し、30分後に耳介厚を測定、抗原を耳介に塗布した。その2時間後に再び耳介厚を測定し、抗原刺激前後の耳介厚の差を浮腫として表した。F-フコイダンにより誘導されたガレクチン9の関与を調べるため、ガレクチン9のアンタゴニストであるラクトースを抗原特異的IgE, Domestic conferenceマコンブ由来F-フコイダンによる新規Ⅰ型アレルギー抑制機構Oral presentation

- 日本学術振興会, 科学研究費助成事業, 基盤研究(C), 神戸学院大学, 01 Apr. 2018 - 31 Mar. 2023乳酸発酵処理が引き起こす豆乳中の成分変化が脂質代謝改善作用に及ぼす影響オルニチンの生体調節機能について、以下の項目について研究を実施した。 (1)肝細胞の脂質蓄積に及ぼすオルニチンの効果:これまで明らかにしてきたヒト肝細胞株HepG2細胞におけるオルニチンの脂肪蓄積抑制効果に関する詳細な機序をAMPKシグナルの下流シグナルとしてオートファジーの促進に着目し、オートファジーの阻害剤であるクロロキンを用いて検討したが、脂肪蓄積抑制効果に影響を及ぼすことはなかった。 (2)肝細胞におけるアンモニア誘導性細胞死に及ぼすオルニチンの保護効果:マウス肝細胞株Hepa1c1c7細胞にオルニチンを作用させると、アンモニア誘導性の細胞死を抑制すると共に、培養液中の尿素含量が上昇する。今年度は、このアンモニアを尿素に解毒することで細胞死を抑制していると考え、オルニチン処理時の尿素回路の亢進について検討した。オルニチンを作用させたHepa1c1c7細胞では細胞中のオルニチン濃度が上昇すると共に、オルニチンの前駆物質アルギニン濃度も増加していたことから、尿素回路が亢進していることが明らかになった。また、ヒト肝細胞株HepG2細胞においてもオルニチン処理により同様のイベントが確認できた。つまり、オルニチンの肝細胞保護機能の機序のひとつとして、尿素回路を亢進することで毒性を示すアンモニア解毒していると考えた。 (3)オルニチン経口摂取時における体内動態:ICRマウス(オス、6週齢)に胃ゾンデを用いて375 mg/Lのオルニチン水溶液200 μLを単回で経口投与し、経時的に血中および肝臓中にオルニチン濃度を調べた。血中、肝臓中のいずれにおいても経時的にその濃度は上昇し、血中では45分後、肝臓中では1時間後に最大濃度に達した。一方、自由摂取ではオルニチンの蓄積は見られなかった。
