- Centromeres are essential for chromosome segregation, yet they are among the most rapidly evolving regions of the genome. The mechanisms driving this rapid evolution of centromeric sequences are still not well understood. In this study, we identified the centromeric sequences of a wheat-infecting Pyricularia oryzae strain (Br48) using CENP-A chromatin immunoprecipitation followed by high-throughput sequencing. The Br48 centromeres range from 71 kb to 101 kb in length and are highly AT-rich (72.1-75.5%) and repeat-rich (63.4-85.0%). These regions are also enriched in H3K9me3 and 5-methylcytosine but depleted of H3K4me2 and H3K27me3. During the analysis of repetitive sequences in the Br48 centromere, we identified a stretch of approximately 530 bp that is tightly associated with centromeres in the Pyricularia fungus. We designated this element as the centromere-associated IR element (CenIR), as it often forms inverted repeat structures with two elements adjacent in reverse orientation. A comparison of putative centromere sequences across phylogenetically distinct P. oryzae strains suggests that changes in centromeric sequences are non-uniform across chromosomes and do not always align with the fungal phylogenetic relationships. - Wheat blast caused by Pyricularia oryzae pathotype Triticum is now becoming a very serious threat to global food security. Here, we report an essential pathogenicity factor of the wheat blast fungus that is recognized and may be targeted by a rice resistance gene. Map-based cloning of Pwt2 showed that its functional allele is the ACE1 secondary metabolite gene cluster of the wheat blast fungus required for its efficient penetration of wheat cell walls. ACE1 is required for the strong aggressiveness of Triticum, Eleusine, and Lolium pathotypes on their respective hosts, but not for that of Oryza and Setaria pathotypes on rice and foxtail millet, respectively. All ACE1 alleles found in wheat blast population are recognized by a rice resistance gene, Pi33, when introduced into rice blast isolates. ACE1 mutations for evading the recognition by Pi33 do not affect the aggressiveness of the rice blast fungus on rice but inevitably impair the aggressiveness of the wheat blast fungus on wheat. - Abstract Wheat blast is an important disease that affects cross-border food security. The causal pathogen, Pyricularia oryzae (syn. Magnaporthe oryzae) Triticum pathotype (MoT), forms a phylogenetic group distinct from the other P. oryzae pathotypes pathogenic on various gramineous plants. Here we developed MoT-specific primer sets for qPCR and LAMP detection methods, and their amplification specificity and efficiency were tested in comparison with previous reported MoT-specific primers. The amplification efficiency and specificity were affected by the polymerase premix. To evaluate the detection specificity, purified DNA samples of 15 MoT isolates, 89 non-MoT isolates of Pyricularia spp., and crude DNA from wheat plants inoculated with MoT were used. - Abstract Horizontal gene transfer (HGT) is a means of exchanging genetic material asexually. The process by which horizontally transferred genes are domesticated by the host genome is of great interest but is not well understood. In this study, we determined the telomere-to-telomere genome sequence of the wheat-infecting Pyricularia oryzae strain Br48. SNP analysis indicated that the Br48 strain is a hybrid of wheat- and Brachiaria-infecting strains by a sexual or parasexual cross. Comparative genomic analysis identified several megabase-scale “insertions” in the Br48 genome, some of which were possibly gained by HGT-related events from related species, such as P. pennisetigena or P. grisea. Notably, the mega-insertions often contained genes whose phylogeny is not congruent with the species phylogeny. Moreover, some of the genes have a close homolog even in distantly related organisms, such as basidiomycetes or prokaryotes, implying the involvement of multiple HGT events. Interestingly, the levels of the silent epigenetic marks H3K9me3 and H3K27me3 in a genomic region tended to be negatively correlated with the phylogenetic concordance of genes in the same region, suggesting that horizontally transferred DNA is preferentially targeted for epigenetic silencing. Indeed, the putative HGT-derived genes were activated when MoKmt6, the gene responsible for H3K27me3 modification, was deleted. Notably, these genes also tended to be up-regulated during infection, suggesting that they are now under host control and have contributed to establishing a fungal niche. - Transposable elements are common targets for transcriptional and post-transcriptional gene silencing in eukaryotic genomes. However, the molecular mechanisms responsible for sensing such repeated sequences in the genome remain largely unknown. Here, we show that machinery of homologous recombination (HR) and RNA silencing play cooperative roles in copy number-dependent de novo DNA methylation of the retrotransposon MAGGY in the fungus Pyricularia oryzae. Genetic and physical interaction studies revealed that RecA domain-containing proteins, including P. oryzae homologs of Rad51, Rad55, and Rad57, together with an uncharacterized protein, Ddnm1, form complex(es) and mediate either the overall level or the copy number-dependence of de novo MAGGY DNA methylation, likely in conjunction with DNA repair. Interestingly, P. oryzae mutants of specific RNA silencing components (MoDCL1 and MoAGO2) were impaired in copy number-dependence of MAGGY methylation. - Eleusine isolates (members of the Eleusine pathotype) of Pyricularia oryzae are divided into two subgroups, EC-I and EC-II, differentiated by molecular markers. A multilocus phylogenetic analysis revealed that these subgroups are very close to Eragrostis isolates. EC-II and Eragrostis isolates were exclusively virulent on finger millet and weeping lovegrass, respectively, while EC-I isolates were virulent on both. The avirulence of EC-II on weeping lovegrass was conditioned by an avirulence gene, PWL1. All EC-II isolates shared a peculiar structure (P structure) that was considered to be produced by an insertion (or translocation) of a DNA fragment carrying PWL1. On the other hand, all EC-I and Eragrostis isolates were noncarriers of PWL1 and shared a gene structure that should have predated the insertion of the PWL1-containing fragment. - BACKGROUND: White root rot disease caused by Rosellinia necatrix is one of the most important threats affecting avocado productivity in tropical and subtropical climates. Control of this disease is complex and nowadays, lies in the use of physical and chemical methods, although none have proven to be fully effective. Detailed understanding of the molecular mechanisms underlying white root rot disease has the potential of aiding future developments in disease resistance and management. In this regard, this study used RNA-Seq technology to compare the transcriptomic profiles of R. necatrix during infection of susceptible avocado 'Dusa' roots with that obtained from the fungus cultured in rich medium. RESULTS: The transcriptomes from three biological replicates of R. necatrix colonizing avocado roots (RGA) and R. necatrix growing on potato dextrose agar media (RGPDA) were analyzed using Illumina sequencing. A total of 12,104 transcripts were obtained, among which 1937 were differentially expressed genes (DEG), 137 exclusively expressed in RGA and 160 in RGPDA. During the root infection process, genes involved in the production of fungal toxins, detoxification and transport of toxic compounds, hormone biosynthesis, gene silencing and plant cell wall degradation were overexpressed. Interestingly, 24 out of the 137 contigs expressed only during R. necatrix growth on avocado roots, were predicted as candidate effector proteins (CEP) with a probability above 60%. The PHI (Pathogen Host Interaction) database revealed that three of the R. necatrix CEP showed homology with previously annotated effectors, already proven experimentally via pathogen-host interaction. CONCLUSIONS: The analysis of the full-length transcriptome of R. necatrix during the infection process is suggesting that the success of this fungus to infect roots of diverse crops might be attributed to the production of different compounds which, singly or in combination, interfere with defense or signaling mechanisms shared among distinct plant families. The transcriptome analysis of R. necatrix during the infection process provides useful information and facilitates further research to a more in -depth understanding of the biology and virulence of this emergent pathogen. - Three ourmia-like viruses, designated Pyricularia oryzae ourmia-like virus (PoOLV) 1 to 3, were identified in a wheat-infecting isolate of P. oryzae. The sizes of the full-length PoOLV1-3 genomes were determined to be 2,528, 1,671, and 2,557 nt. Interestingly, we also found two abundant single-stranded RNAs sharing their 5' terminal 25 and 255 nt with PoOLV1 RNA and PoOLV3 RNA, respectively. The PoOLV1- and PoOLV3-associated RNAs (ARNA1 and ARNA3) were 639 and 514 nt in length, and possessed one and two small ORFs, respectively. In the field isolates of P. oryzae, PoOLVs and ARNAs were detectable at varying levels, and the levels of PoOLV1 and ARNA1 as well as those of PoOLV3 and ARNA3, were tightly correlated. In addition, gene silencing of PoOLV1 and PoOLV3 resulted in a reduction of ARNA1 and ARNA3, respectively. - Pyricularia is a fungal genus comprising several pathogenic species causing the blast disease in monocots. Pyricularia oryzae, the best-known species, infects rice, wheat, finger millet, and other crops. As past comparative and population genomics studies mainly focused on isolates of P. oryzae, the genomes of the other Pyricularia species have not been well explored. In this study, we obtained a chromosomal-level genome assembly of the finger millet isolate P. oryzae MZ5-1-6 and also highly contiguous assemblies of Pyricularia sp. LS, P. grisea, and P. pennisetigena. The differences in the genomic content of repetitive DNA sequences could largely explain the variation in genome size among these new genomes. Moreover, we found extensive gene gains and losses and structural changes among Pyricularia genomes, including a large interchromosomal translocation. We searched for homologs of known blast effectors across fungal taxa and found that most avirulence effectors are specific to Pyricularia, whereas many other effectors share homologs with distant fungal taxa. In particular, we discovered a novel effector family with metalloprotease activity, distinct from the well-known AVR-Pita family. We predicted 751 gene families containing putative effectors in 7 Pyricularia genomes and found that 60 of them showed differential expression in the P. oryzae MZ5-1-6 transcriptomes obtained under experimental conditions mimicking the pathogen infection process. - Small RNA (sRNA)-mediated gene silencing phenomena, exemplified by RNA interference (RNAi), require a unique class of proteins called Argonautes (AGOs). An AGO protein typically forms a protein-sRNA complex that contributes to gene silencing using the loaded sRNA as a specificity determinant. Here, we show that MoAGO2, one of the three AGO genes in the fungus Pyricularia oryzae (Magnaporthe oryzae) interferes with RNAi. Gene knockout (KO) studies revealed that MoAGO1 and MoAGO3 additively or redundantly played roles in hairpin RNA- and retrotransposon (MAGGY)-triggered RNAi while, surprisingly, the KO mutants of MoAGO2 (Δmoago2) showed elevated levels of gene silencing. Consistently, transcript levels of MAGGY and mycoviruses were drastically reduced in Δmoago2, supporting the idea that MoAGO2 impeded RNAi against the parasitic elements. Deep sequencing analysis revealed that repeat- and mycovirus-derived small interfering RNAs were mainly associated with MoAGO2 and MoAGO3, and their populations were very similar based on their size distribution patterns and positional base preference. Site-directed mutagenesis studies indicated that sRNA binding but not slicer activity of MoAGO2 was essential for the ability to diminish the efficacy of RNAi. We conducted laboratory experiments to assess the disease control effects of pH-decreasing fertilizer as a new control material for lettuce big-vein disease. Results show that the occurrence of big-vein disease was suppressed in concomitance with the descent of soil pH by applying the fertilizer.

- Pseudomonas fluorescens FPT9601株は,トマトにおいて植物成長促進根圏細菌として働き,ナス科植物青枯れ病菌に対する抵抗性を付与することで単離同定された。本菌はモデル植物であるシロイヌナズナにおいても,誘導全身抵抗性を引き起こす。シロイヌナズナのサリチル酸(SA),ジャスモン酸(JA)及びエチレン(ET)経路の変異株を用いて調査を行った所,FPT9601株が誘導する病害抵抗性にJAおよびET経路の関与が示唆された。次に約23.000遺伝子のプローブセットによるマイクロアレイ解析を行った結果,2倍以上の発現変動で上昇した遺伝子が95個,低下したものが105個であった。その中にオーキシン応答性遺伝子の発現増高が認められ,本菌による植物の成長促進はオーキシンの作用であることが示唆された。この他に二次代謝経路に関わる遺伝子の誘導や抑制,タンパク質合成系遺伝子の抑制などの傾向が認められた。また根粒菌感染で発現が増高することから命名されたノジュリン遺伝子群に含まれる遺伝子の誘導なども見られた。マイクロアレイ解析の結果,FPT9601感染により発現が上昇していた遺伝子のトランスポゾン挿入変異体におけるFPT9601株による誘導全身抵抗性について調査した所,転写因子などいくつかの変異体において,誘導抵抗性の低下が認められた。Japanese Society of Soil Microbiology, 2012, Soil Microorganisms, 66(2) (2), 43 - 46, Japanese

- Transcriptional activity of the retrotransposon OARE-1 was monitored in oat leaves infected with pathogenic fungi. The transcription of OARE-1 was upregulated in both incompatible and compatible interactions between an oat cultivar and Magnaporthe grisea isolates but more extensively in the compatible. OARE-1 was also strongly activated in a Pc2/Vb-carrying oat line inoculated with Helminthosporium victoriae. The upregulation in the Pc2/Vb-carrying oat line was reproduced by treatment with victorin C, the major compound of the host-specific toxin produced by the fungus. These results suggest that OARE-1 is responsive to various signals or stresses associated with compatibility and incompatibility.Phytopathological Society of Japan, 2002, J. Gen. - The complete nucleotide sequence of a set of cDNA clones designated KU1 strain, from which infectious in vitro transcripts can be transcribed, of the ATCC66 strain of brome mosaic virus (BMV) was determined and compared with the Russian strain of BMV in both nucleotide and the deduced amino acid sequence. Each corresponding genomic RNA in the KU1 strain and the Russian strain was homologous by more than 97% in nucleotide sequence. Base substitution of the transition type was more frequent than that of the transversion type in all three genomic RNAs. Amino acid changes in three regions where significant amino acid sequence homology is shown among the Sindbis-like plant RNA viruses were mostly substitutions for amino acids categorized to similar groups. The nucleotide sequence of the 5' noncoding region in RNA1 and RNA3 was identical in the corresponding RNAs of the two strains while seven base changes were observed in RNA2. Any base change in the 3' noncoding region did not disrupt a predicted secondary structure. Two adjacent adenine residues were absent in the noncoding leader sequence of the coat protein gene of both KU1 strain and ATCC66 strain. - カリフラワーモザイクウイルス(CaMV), CM1841系統は, 昆虫媒介性に関与するウイル遺伝子(ORF II)にアミノ酸置換がある非昆虫媒介性のCaMV分離株として知られてきた。CM 1841感染葉では, ORF II産物(p18)蓄積量が昆虫媒介性のCaMVと比較して少ないが, 病徴発現初期には昆虫媒介性のCaMV系統の25〜50%程度のp18が蓄積している (Nakayashiki et al. J. Gen. - RNA Silencing in a Phytopathogenic Fungus, Magnaparthe griseaRNAサイレンシングは動物,植物,および糸状菌などの幅広い真核生物が持つ外来核酸に対する防御機構と考えられている.我々はコムギから分離されたいもち病菌Br48にGFP遺伝子を導入し,強いGFP緑色蛍光を示す株を得た.この形質転換体にGFP RNAをセンス,アンチセンス,ヘアピンなどの様々な形で転写するコンストラクトをそれぞれ導入し,GFP蛍光の増減を調査した結果,ヘアピン構造をとるGFP RNAを転写するコンストラクトが高頻度でGFP蛍光の抑制を誘導することが明らかとなった.これらGFPの発現が抑制された株ではGFP mRNAの蓄積が顕著に低下していることがノーザン解析により示された.また,GFPのサイレンシングが観察された株ではGFPヘアピンRNAに相当する3.2kbのバンドが常に観察され,両者の間に相関が認められた.以上の結果は,いもち病菌にRNAサイレンシング機構が存在することを示し,ゲノム解析が進展している本菌の逆遺伝学的な遺伝子解析に有用な手法となることが期待される.The Phytopathological Society of Japan (PSJ), 25 Feb. 2003, Annals of the Phytopathological Society of Japan, 69(1) (1), 51 - 51, Japanese
- Analysis of Apoptosis Induction Associated with Induced Resistance in Oat. : 13) Cytological Analysis of Calciumregulated Reactive Oxygen Species Generation and Cell Death植物の抵抗性発現およびアポトーシスの誘導にCa^<2+>およびROSは重要な役割を担っている.シロイヌナズナの細胞膜在性NADPHオキシダーゼAtrbohはCa^<2+>結合部位を有することが報告され,Ca^<2+>とROS生成には深い相関性があることが明らかとなっている.しかし現在までにCa^<2+>に制御されるROS生成の詳細な細胞学的解析は行われていない.そこで本研究ではカルシウムイオノフォアA23187がエンバク葉肉細胞に誘導する細胞死およびROS生成についての解析を行った.A23187処理したエンバクでは,明瞭なDNAラダー,グロマチンの凝集が認められ,EM-TUNELポジティブであり,アポトーシス様の細胞死が起こっていることが示唆された.塩化セリウム法により,細胞膜およびミトコンドリア,葉緑体,パーオキシゾームなどの細胞内小器官で強いH_2O_2の蓄積が認められた.NADPHオキシダーゼ阻害剤であるDPIを供処理すると,細胞膜でのH_2O_2蓄積および細胞死が抑制されたが,これに伴い細胞内小器官でのH_2O_2蓄積も抑制された.このことより,細胞膜でのO_2-生成が細胞内小器官での二次的なROS生成を誘導することが示唆された.The Phytopathological Society of Japan (PSJ), 25 Feb. 2003, Annals of the Phytopathological Society of Japan, 69(1) (1), 55 - 55, Japanese

- Involvement of gacA Gene in the Suppression of Tomato Bacterial Wilt by Pseudomonas fluorescens FPT9601 :Pseudomonas fluorescens FPT9601, a plant growth-promoting rhizobacterium (PGPR) isolated from tomato rhizosphere, can protect tomato (Lycopersicon esculentum Mill) from bacterial wilt disease caused by Ralstonia solanacearum. This strain produces antibiotics 2,4-diacetylphloroglucinol (2,4-DAPG) and hydrogen cyanide (HCN). It also produces proteases and uncharacterized siderophores (Sid). A mutant strain SM2214, obtained by Tn5 insertion, did not produce 2,4-DAPG, HCN or proteases, but overproduced Sid. Marker-exchange mutagenesis confirmed that a single transposon insertion caused the multiple phenotypic changes of this mutant. Complementation of the mutant with a 1.3-kb DNA fragment that was amplified from genomic DNA of the wild-type P. fluorescens strain by PCR could restore the lost functions of the mutant strain. Nucleotide sequencing revealed that the fragment contained a 642-bp open reading frame (ORF) highly homologous to the regulator responser gene gacA. The in vitro anti-bacterium test and plant protection experiment under greenhouse conditions indicated that the gacA gene played an important role in the suppression of tomato bacterial wilt disease. - Signal Mediators for Phytoalexin Production in Defense Response of Oats Elicited by Victorin as a Specific Elicitor :Signals mediating phytoalexin(PA)production were analyzed in primary leaves of oats cv.Iowa X469 treated with an elicitor victorin. Production of the PA avenanthramide A was inhibited by DPI, an inhibitor of NADPH oxidase/nitric oxide synthase(NOS)and the NOS inhibitors L-NMMA and 1, 3-PBIT. However, catalase and superoxide dismutase (SOD)hardly suppressed it. From the data, NO functions as a major reactive oxygen species in signal transduction leading to PA production in the defense response of oats. EGTA, verapamil and ruthenium red inhibited PA production, suggesting that Ca^<2+> influx into the cytoplasm and intracellular Ca^<2+> movement are involved in the defense response. Trifluoperazine, a calmodulin function inhibitor, and K-252a, a serine/threonine kinase inhibitor, also suppressed the accumulation, whereas okadaic acid, a serine/threonine phosphatase inhibitor, did not suppress it, suggesting the involvement of calmodulin and protein kinase, but not of phosphatase in PA production.Phytopathological Society of Japan, 2000, Journal of general plant pathology : JGPP, 66(3) (3), 185 - 190, English
- Pathogenicity, Mating Ability and DNA Restriction Fragment Length Polymorphisms of Pyricularia Populations Isolated from Gramineae, Bambusideae and Zingiberaceae Plants :Eighty-five Pyricularia isolates were collected from 29 host species of Gramineae, Bambusideae and Zingiberaceae plants sampled in Brazil, Uganda, Ivory Coast, India, Nepal, China, Indonesia and Japan.These isolates were compared on the basis of pathogenicity, mating ability and restriction fragment length polymorphisms with single-copy DNA probes.Based on the pathogenicity to eight differential gramineous plants, these isolates were classified into seven pathotypes:finger millet type, foxtail millet type, common millet type, rice type, crabgrass type, Italian ryegrass / weeping lovegrass type, and non-cereal / grass type.Genetic variation among these isolates was assessed by RFLP analysis with two restriction enzymes and nine single-copy DNA probes isolated from a finger millet strain.An UPGMA dendrogram based on the RFLPs revealed that the 85 isolates could be classified into seven major groups.Isolates from cereal crops(finger millet, foxtail millet, common millet, wheat and rice)and a grass, Brachiaria plantaginea, were clustered into a single group.They were further divided into six subgroups corresponding to the pathotypes.Among cereal crop isolates only an isolate from pearl millet was located into a different group.The remaining isolates were clustered into five groups designated as the crabgrass group, the buffelgrass and jungle rice group, the rice cutgrass, knotroot bristlegrass and Setaria tomentosa group, the bamboo and bamboo grass group and the Zingiber mioga group.The isolates from cereal crops were generally capable of mating with finger millet strains and constituted a closed mating compatibility group.These results suggested that the isolates from cereal crops from a single group with a common ancestor although they are pathogenic to taxonomically diverse plants.A combined analysis of the pathogenicity and genetic similarity suggested that the transmission of M.grisea isolates occurs in natural agroecosystems between finger millet and Eleusine africana, goosegrass or Bambusa arundinacea, between foxtail millet and green bristlegrass, and between rice and tail fescue, Italian ryegrass, sweet vernalgrass, reed canarygrass or Oryza longistaminata.Phytopathological Society of Japan, 2000, Journal of general plant pathology : JGPP, 66(1) (1), 30 - 47, English

- Genetic Diversity in Pyricularia Isolates from Various Hosts Revealed by Polymorphisms of Nuclear Ribosomal DNA and the Distribution of the MAGGY RetrotransposonPyricularia isolates from various gramineous phants were subjected to RFLP and sequence analyses of rDNA to clarify their genetic relationships. Based on rDNA-RFLP patterrns produced by digestion with two restriction enzymes, 71 isolates from 27 plant species were grouped into 13 rDNA types. To estimate their genetic relatedness, a UPGMA dendrogram was constructed based on the rDNA-RFLP profiles. The ITS2 regions of 13 representative isolates were also amplified by PCR and directly sequenced to construct another dendrogram. In both dendrograms, isolates from agronomically important crops (rice, foxtail millet, common millet, finger millet, wheat and oats) and their relatives were classified into a single cluster. Isolates from weeds, such as crabgrass and buffelgrass, were located outside the crop isolate cluster, and Japanese bamboo or bamboo grass isolates were farthest from the crop isolate cluster. In surveying the distribution of a retrotransposon, MAGGY, in these isolates, MAGGY was present in multiple copies in isolates from rice, foxtail millet and six other hosts, which formed a single subcluster within the crop isolate cluster. Multiple copies of MAGGY were also detected in an isolate from buffelgrass, which was distant from the other MAGGY carriers in ther DNA dendrograms. - Molecular Analysis of the Wheat Blast Population in Brazil with a Homolog of Retrotransposon MGR583We have identified a family of dispersed repetitive DNA sequences in the genome of a Magnaporthegrisea isolate from finger millet (Eleusine coracana). Southern blot analyses showed that this element is present in a moderate copy number (30-40 copies) in the genome of blast isolates from wheat (Triticum aestivum). DNA sequence data suggested that this element contains a region highly homologous to the reverse transcriptase domain of MGR583, a poly A-type retrotransposon. Using the reverse transcriptase domain of this element as a molecular probe, the genetic structure of the wheat blast population was examined. - Population Structure of the Rice Blast Fungus in Japan Examined by DNA FingerprintingDNA fingerprinting with multilocus probes, MAGGY and MGR586, was conducted to investigate the population structure of the rice blast fungus Magnaporthe grisea in Japan. Two-hunredseventy-eight field isolates collected during 1993-1997 and 22 archival isolates collected in 1976 from various cultivars and locations were examined. Two fingerprint groups were identified at the 70% similarity level in each collection. A one-to-one correspondence was found between the groups in field isolates and the groups in archival isolates. These result ssuggest that two discrete lineages exist in the rice blast population. They were temporarily designated as JL1 and JL2 (Japanese Lineage). JL1 was a predominant lineage representing 97% and 77% of the field and archival isolates, respectively, and present through out Japan. In addition, these two lineages corresponded to two of the five lineages previously detected in collections made before 1960, suggesting that the structure of the rice blast population in Japan had drastically changed during 1960-1976. The two lineages showed a very similar virulence spectrum, and no relationship could be recognized between lineages and pathotypes. - Replication of Cauliflower Mosaic Virus ORF I Mutants in Turnip ProtoplastsWe succeeded in infecting turnip protoplasts with a cloned cauliflower mosaic virus (CaMV) DNA, pCa122, which contains 1.2 copy of CaMV genomic DNA and a plasmid expressing open reading frame (ORF) VI products (pEXP6) using polyethylene glycol. Fluorescent antibody staining showed that up to 50% of protoplasts were infected. It was difficult to detect the progeny DNA and the viral protein in protoplasts inoculated with pCa122 alone. Co-transfection with the plasmid pEXP6 produced larger fluorescing specks in each infected protoplasts and increased the accumulation of the progeny DNA and some other viral proteins to detectable levels. Using this protoplast system, three CaMV ORF I insertional mutants which were not infectious on turnip plants were tested for their infectivity on turnip protoplasts. Viral DNA and products accumulated in infected protoplasts to the same extent of the wild type DNA-infected protoplasts. - Aphid transmissible Japanese isolate of cauliflower mosaic virus M (CaMV-M) was cloned into pUC19 and its ORF II region was sequenced. Comparison of this sequence with that of CM1841 (non-aphid transmissible isolate of CaMV) showed that there were nine base changes, four of which gave rise to alterations in the predicted amino acid sequence. The novel sRNAs were characterized by its smaller size (16-18nt), Dicer- and/or RdRP-independent biogenesis, and nonbinding to AGO protien. Northern blotting analysis revealed several sizes of precursors of the novel sRNA, indicating multiple steps of processing for its biogenesis. Consistently, RNA-seq analysis suggested that some of the precursors were generated via complex splicing. GFP reporter assay indicated that the novel sRNA was not functional in the gene silencing pathways, supporting the idea that these sRNAs possess so-far unknown biological roles.

Consequently, a great progress has been made, leading to breakthroughs: 1) Discovery of RNAi mediated antiviral defense that requires Dicer but not AGO in Cryphonectria parasitica, 2) Detection of inhibitory effects on RNA silencing activities against RNA and DNA viruses, and transgenes in Pyricularia oryzae, 3) Development of a screening protocol for host factors required for virus perception through transcription induction of key genes of RNAi upon virus infection in C. parasitica, 4) Identification of the SAGA complex as a transcriptional regulator of fungal RNA silencing. These findings will bring about a paradigm shift in RNAi. Fungi are now established as the third position next to animals and plants in antiviral defense research. Note that most of the obtained data were published in international, high-quality journals.

- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (A), Kobe University, 2002 - 2004Identification and cloning of genes conditioning species-specific parasitism of Magnaporthe oryzaeTo clarify genetic mechanisms of plant-species specificity of Magnaporthe oryzae, we crossed a Triticum isolate with a Setaria isolate, an Avena isolate, and an Oryza isolate. Using the F_1 progenies derived from these crosses, we identified PWT1, PWT2 (conditioning avirulence of the Setaria isolate on wheat), PWT3, PWT4 (conditioning avirulence of the Avena isolate on wheat), PWT5 (conditioning avirulence of the Oryza isolate on wheat), PFM1, PFM2 (conditioning avirulence of the Triticum isolate on foxtail millet), and PAT1 (conditioning avirulence of the Triticum isolate on oat). We also identified a resistance gene corresponding to PWT4, and designated it as Rwt4. Rwt4 corresponded to PWT4 in a one-to-one manner, suggesting that the plant species specificity of the Triticum/Avena isolates on wheat is conditioned by gene-for-gene interactions. From these avirulence genes, PWT1, PWT4, and PAT1 were chosen as targets for molecular cloning. Using bulked segregant analysis, we found molecular markers that are closely linked to PWT1, PWT4 and PAT1.

Deletion analysis of the MAGGYLTR promoter Several constructs having a deletion in the MAGGY promoter were made and examined for the promoter activity upon stress using the GUS gene as a reporter. The deletion constructs with the GUS gene were introduced into M.grisea isolate Br 48. Total protein was extracted from the M.grisea transformants after beat shock treatment (42C, 1hr), and the GUS activity was measured using MUG as a substrate. The construct that lacks up to 60 bp at 5' terminus of LTR retained the promoter activity and responded to heat shock whereas one lacking 90 bp at 5' terminus lost the promoter activity. The results indicated that some critical sepuence required for both promoter activity and stress response occur in the 60 to 90 region of MAGGYLTR. 2. Analysis of the promoter activity of MAGGY during infection processes using the GFP gene To investigate the promoter activity of MAGGY during infection processes, GFP was expressed in a M.grisea transformant under the control of the MAGGY promoter. Infection test was performed with the GUS-expressing transformants. However, no obvious upregulation of the MAGGY promoter was found during infection processes on either resistant or susceptible cultivar of rice. 3. Isolation of a new LTR-retrotransposn from M.grisea. As a new candidate for a stress responsive transposable element, an LTR-retrotransposon, Pyret was isolated from M.greisea. Pyret containg gag and pol genes as do ordinary gypsy-class retrotransposons. However, this element is unique since it carries an extra domain in the gag gene, which is designated as WCCH domain according to a characteristic amino acid sequence. The WCCH domain was found only in four gypsy-class retrotransposons identified in fungal genomes so far. Therefore, the functional role of the WCCH domain is unknown at this moment.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (A), Kobe University, 1999 - 2001Cooperative research on biologicalcontrolofplantpestCurrent status in China of biological control of plant diseases and insect pest was studied. The recent development of biological research and application was summarized in the final report. We especially studied on biological control of tomato bacterial wilt by PGPR strain, Pseaudomonas fluorescence FPT9601. It was found that the strain could colonize within the root tissues of both monocot and dicot plants. They formed microcolonies in a concave portion in the root surface such as injured places from which bacterial pathogens can be easily invaded, indicating that the presence of PGPR strains in the portion seem to be appropriate for suppressing the pathogens. Once the bacteria were invaded in roots, an induced resistance was activated in plants. Flaggellin components of the bacteria were important factor for colonization and induction of defense response in plants. Also, 2,4-DAPG, diacetylphlorogrucinol, was found as a major antibacterial factor to suppress the bacteria wilt strain. On the other hand, the methods employed for biological control of insect pests have been studied in China. It was found that introduction of natural enemies, conservation, environment management for manipulation of natural enemies, augmentation and release of biological control agents have been developed and they greatly contribute to the improvement of the biological control in China.
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B)., KOBE UNIVERSITY, 1998 - 2000Molecular analysis of species-specific parasitism in Magnaporthe griseaMechanisms of the species-specific parasitism in Magnaporthe grisea were investigated. An F1 population derived from a cross, a Setaria isolate x a Tritucum isolates, showed bifactorial segregation in the pathogenicity on wheat. These genes were designated as Pwt1 and Pwt2. Another F1 population derived from a cross, an Avena isolate and a Triticum isolate, showed monofactorial segregation on oat. This gene was designated as Pat1. Closely linked AFLP markers were found in bulked segregant analyses. Chromosome walking is under way toward cloning of those genes. A phytotoxin was extracted from a Digitaria isolate. The toxin was revealed to be Pyrichalasin H.Among various gramineous plants, crabgrass (the host of Digtaria isolates) showed the highest susceptibility to this toxin. Pyrichalasin H was produced in a large quantity only by Digitaria isolates, and Pyrichalasin H-defidient mutants lost their pathogenicity on crabgrass. Crabgrass leaves treated with Pyrichalasin H became accessible to nonpathogens. These results suggested that Pyrichalasis H is a determinant of the species-specific parasitism of Digitaria isolates. Retrotransposon MAGGY was the most active element among transposable elements that have been found in the genome of Magnaporthe grisea. MAGGY was present in Oryza isolates and Setaria isolates, but absent in other isolates. We assuned that this element may have been involved in the evolution of the race- cultivar specificity in those isolates.
- 日本学術振興会, 科学研究費助成事業, 基盤研究(C), 神戸大学, 1998 - 1999イネいもち病菌の病原性変異機構の解析−特に転移因子の関与についてイネいもち病菌に存在するレトロトランスポゾンMAGGYの、同菌の病原性変異への関与を調査する目的で、今年度は二項目の研究を行い以下に記した結果を得た。 1. MAGGYの活性化条件を調べるために、MAGGYのプロモーターが存在するLTR配列の下流にレポーターとしてGUS遺伝子を結合したプラスミドを作製した。このプラスミドをPEG法によりイネいもち菌に導入して、ストレスによるMAGGYの活性化を検討した。その結果、MAGGYのプロモーター活性は、42℃、1時間の熱ショックにより、約60倍増高することが明らかになった。また、500mM過酸化水素処理によっても約20倍の増高が認められた。今後、このコンストラクトを用いることによって、菌の感染行動と関連したMAGGYの活性化についてさらに調査を続ける予定である。 2. 通常感染が成立しないイネ-いもち病菌の組み合わせでも、イネの穂ばらみ期に注射接種することによって低頻度の感染が起こることが報告されている。この条件下で、感染が起こった病斑から単胞子分離を行い、これらの菌系における病原性の変異とMAGGYの転移との相関性を調査した。これまでに約170菌系を単胞子分離により確立し、その内の15菌系において病原性の変化が見られた。現在の所、それと明確に対応するMAGGYの転移バンドは確詔されていないが、今後F1菌系における転移バシドと病原性の分離と連鎖を調査し、更に解祈を進める予定である。
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Kobe University, 1998 - 1999Rapid identification of races and species-specific pathotypes using DNA-fingerprintingDNA fingerprinting with miltilocus probes, MAGGY and MGR586, were conducted to investigate the population structure of the rice blast fungus, Magnaporthe grisea in Japan. Analyses of ca. 300 isolates revealed that two discrete lineages exist in the current rice blast population in Japan. They were designated as JL1 and JL2. There two lineages corresponded to two of the five lineages previously detected in collections made before 1960, suggesting that the structure of the rice blast population in Japan had drastically changed during 1960-1976. Rice blast population in Vietnam were also subjected to DNA fingerprinting with MAGGY probe and the pathotype analysis. These analyses revealed that the blast population in the northern region of Vietnam is genetically more diverse than that in the southern region. In both populations no relationships were detected between lineages and races, suggesting that the "lineage exclusion hypothesis" is not applicable to those populations. DNA fingerprinting experiments were carried out for establishing a system of DNA-diagnosis of six host-specific subgroups of M. grisea ; Oryza isolates, Triticum isolates, Panicum isolates, Setaria isolates, Eleusine isolates, and Digitaria isolates. We propose the following formulas for distinguishing those subgroups. (1)HindIII+MGR583 : to identify each group (2)SalI+MAGGY : to identify the Oryza and the Setaria isolates (3)EcoRV+MGR583 : to identify the Triticum isolates (4)HindIII+Pot2 : to identify the Digitaria isolates
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Kobe University, 1996 - 1997Molecular Mechanisms of Resistance Gene Expression in the Crown Rust of OatsThe purpose of this research project is to elucidate the molecular mechanisms of recognition and signal transduction which regulate disease occurence in oats and crown rust interactions. Special reference was concerned with (1) a role of release of activated oxygen, (2) cloning of genes activated during resistance expression, such as chitinase as PR protein and genes responsible to phytoalexin production and signal transduction. As a result, it was found that evolvement of activated oxygen is absolutely required for initiation of genes for phytoalexin production. The cells affected by infection as well as elicitors reacted positively to stains which detect free radicals including activated oxygen. Activated oxygen could be important for induction of resistance as a secondary messengers and induce gene activation for PR protein, phytoalexin and so on. Chitinase was induced in parallel with resistance expression. The enzymes were purified and N-teminal sequence of amino acid was determined and the genes encoded with chitinase have been cloned. In addition, it was found that the chitinases can release chito-oligosaccharides active as elicitor. Finally, it was found that DNA-laddering as a typical symptom of apoptosis was detected in association of resistance response in oat cells treated with rust races and elicitors. Relationships between apoptosis and defense response was confirmed.
- 日本学術振興会, 科学研究費助成事業, 奨励研究(A), 神戸大学, 1994 - 1994いもち病菌の病原性、寄生性分化と連鎖するDNAマーカー配列の検索いもち病菌には各種イネ科植物、あるいは品種に特異的に病原性を持つ種分化やレース分化が起きており、これらの菌は病原性、交配能、小分生子形成能等に異なった生物学的特徴を持っている。これらの生物学的な特徴と連鎖し、それらを担った遺伝子のマーカーとなるDNA配列を検索することを目的とした。本年度はその第一歩としてRFLP分析を行い、それによるいもち菌の系統発生的な解析を行った。 イネ科栽培植物および雑草に寄生性を示す43種の各種いもち病菌から抽出したDNAを、22種のプローブ、2種の制限酵素を用いてRFLP分析した結果、各菌はミョウガ菌群、メヒシバ菌群、栽培植物菌群に大別され、さらに栽培植物菌群は、大きくコムギ・シコクビエ菌群とアワ菌・キビ菌・イネ菌群の2つに分かれることが明らかとなった。各種雑草菌では、ハルガヤ、クサヨシ、ネズミムギ、ヒロハノウシノケグサ等の菌がイネ菌に、ハイキビ菌がキビ菌に、エノコログサ菌がアワ菌に、またトウジンビエ、エゾノサヤヌカグサ、ブッフェルグラス菌等がメヒシバ菌に、それぞれ近縁な菌であることが明らかとなり、雑草菌が新たな菌群を単独で形成することはなかった。これらの結果は、これまでに分かっている各菌の病原性の分化によるグル-ピングとよく一致しており、DNAを用いた病原性遺伝子へのアプローチが有効な方法であることが示された。
