- Abstract Background Food by-products, such as corn germ meal from starch processing, are increasingly used as sustainable feed supplements, reducing competition between food and feed and supporting the valorisation of food waste. However, their effects on gut microbial metabolism and host health remain unclear. This study aimed to determine how corn germ meal fermentation influences microbial community structure and metabolite production using an ex vivo pig faecal culture system. Results Corn germ meal supplementation significantly altered the microbial composition, increasing diversity and enriching fibre-degrading Prevotellaceae, a key bacterial family involved in complex carbohydrate metabolism. Metabolomic analysis revealed marked increases in tryptophan-derived metabolites, including indoleacrylic acid, indolepropionic acid, and indolelactic acid, which act as ligands for the aryl hydrocarbon receptor and have anti-inflammatory properties. Prevotella -mediated catabolite repression reduced Escherichia coli –derived indole formation, redirecting microbial tryptophan metabolism toward the production of these bioactive compounds. Microbial and metabolic responses differed among farms, reflecting farm-specific microbiome structures. Conclusions Corn germ meal supplementation reshapes gut microbial communities, enhances metabolic activity, and promotes the generation of bioactive tryptophan metabolites with potential immunomodulatory effects. These findings highlight the value of corn by-products as dietary fibres that can drive beneficial microbial cross-feeding and influence host intestinal homeostasis. Although demonstrated in an ex vivo setting, this study provides a mechanistic basis and preclinical evidence for future in vivo studies, supporting the sustainable utilisation of food industry by-products to improve gut health and resource efficiency in livestock production.Springer Science and Business Media LLC, Feb. 2026, Journal of Animal Science and Biotechnology, 17(1) (1)[Refereed]Scientific journal

ABSTRACT Interleukin-18 (IL-18), which was originally called gamma interferon (IFN-γ)-inducing factor, has been shown to play an important role in innate and acquired immune responses. In this study, attenuatedErysipelothrix rhusiopathiae strains were engineered to produce porcine IL-18 (poIL-18) and evaluated for their potential immunostimulatory effect in animals. Recombinant poIL-18 was successfully expressed in the recombinantE. rhusiopathiae strains YS-1/IL-18 and KO/IL-18. The culture supernatant of YS-1/IL-18 was confirmed to induce IFN-γ production in murine splenocytesin vitro , and this production was inhibited by incubation with anti-poIL-18 monoclonal antibodies. Furthermore, more IFN-γ production was induced upon stimulation of splenocytes with concanavalin A for splenocytes from mice that were intraperitoneally inoculated with YS-1/IL-18 than for splenocytes from control mice inoculated with the parent strain YS-1. Peritoneal macrophages from mice preinoculated with YS-1/IL-18 exhibited enhanced phagocytosis ofSalmonella enterica subsp.enterica serovar Typhimurium compared with peritoneal macrophages from control mice preinoculated with YS-1. We also confirmed the immunostimulatory effect on humoral immune responses against antigens ofE. rhusiopathiae andMycoplasma hyopneumoniae in gnotobiotic pigs that were orally preinoculated with KO/IL-18. ABSTRACT We have developed a system in which a foreign antigen is delivered and expressed on the surface of an attenuated strain ofErysipelothrix rhusiopathiae YS-1 and have examined the ability of a such recombinantE. rhusiopathiae strain to function as a mucosal vaccine vector. The C-terminal portion, including two repeat regions, R1 and R2, of the P97 adhesin ofMycoplasma hyopneumoniae strain E-1 was successfully translocated and expressed on theE. rhusiopathiae YS-1 cell surface after it was fused to SpaA.1, a cell surface protective antigen ofE. rhusiopathiae. BALB/c mice subcutaneously immunized with theE. rhusiopathiae recombinant strains developed specific antibodies against SpaA.1 protein and were protected from lethal challenge with the highly virulent homologousE. rhusiopathiae Fujisawa-SmR strain, showing the efficacy of this heterologous-antigen expression system as a vaccine againstE. rhusiopathiae infection. To determine whether protective immune responses are induced in target species, newborn, specific-pathogen-free piglets were immunized intranasally with a recombinant strain designated YS-19. The immunized piglets developed specific anti-SpaA.1 immunoglobulin G (IgG) antibodies in their serum and were protected from death by erysipelas, showing that mucosal vaccination of piglets with YS-19 induces systemic immune responses. Furthermore, YS-19-immunized piglets showed higher levels of P97-specific IgA antibodies in the respiratory tract than did YS-1-immunized piglets. - This study was carried out to determine the effects of crowding on weight gain, immune performance and the concentration of salivary stress markers in growing-finishing pigs. Space allowances were 0.37 m2/pig (crowding stress group, n=14) and 0.87 m2/pig (control group, n=6) in a fully slatted facility (5.24 m2). Initial weights averaged 52.3 kg, and the pigs were tested for 14 days. The average daily gains were significantly lower in the crowding stress group than in the control group on days 0-7 (P<0.05) and days 7-14 (P<0.01). The lymphocyte proliferation indexes in response to Concanavalin A were significantly higher in the crowding stress group than in the control group on day 14. The CD4:CD8 ratio was significantly lower in the crowding stress group than in the control group on day 14. CD8+ and CD14+ lymphocytes were significantly higher (P<0.05) in the crowding stress group than in the control group on day 14. Salivary stress markers (Cortisol, IL-18 and IgA) did not change as a result of crowding stress. - Respiratory syndromes and diarrhea among weaned pigs decrease number of marketed pigs and reduce growth performance. This clinical trial was conducted to evaluate the effects of environmental improvements and probiotic supplement on mortality, body weight gain and saliva cortisol levels among weaned pigs reared in a farm, which known to have adversed sanitary conditions. In October 2010, 140 pigs weaned in this month were classified into 4 body weight groups (heavy, medium, light, extreme light) (n=35 each). Each group was further randomly allocated into 4 groups such as reared in; A) cleaned room with probiotics (Lactobachillus casei I-5 (1%)), B) cleaned room without probiotics, C) uncleaned room with probiotics, D) uncleaned room without probiotics. Therefore, 16 pens (8-10 animals each) were allocated into 4 treatment groups (A to D) and 4 blocks by their weights. Body weights of the targeted animals were measured weekly until 9 weeks and saliva obtained at 0, 1, 4 and 8 weeks post weaning were used for cortisol measurement. In extreme light weight group, pigs in cleaned room and those with probiotics had significantly higher body weight gain compared to other groups. In the same weight group, pigs without probiotics had higher mortality compared to other group and this was more evident among castrated male group. At 4 weeks post weaning, cortisol concentration in saliva remained highest levels among castrated male in extreme light weight group without probiotics compared to other groups. From this study, cleaning process and probiotic supplementation were effective on body weight gain and improvement of mortality only for pigs which were lowest body weight group at weaning. - In 2009, a swine farm which is known to have high mortality of growing pigs was used for this clinical trial. Ear tags were attached to 105 piglets which were borne in June, 2009 and allocated into 2 nursing houses, house A with relatively good environmental conditions and house B with bad sanitary condition with high mortality. The house B had 12 time dust concentrations, 4 times ammonia concentration, 8 times nitrous oxicide and 2 times methane concentration higher than those measured in house A. The IgA concentration in the saliva of the pigs raised in house B was 4 times higher than those in house A. Total mortality after weaning in this trial was 26.2% (16/61) and 40.4% (17/42) for growing pigs raised in house A and B, respectively. - Immune responses to the Mycoplasma hyopneumoniae P97 antigen in pigs vaccinated with Erysipelothrix rhusiopathiae YS-19Mycoplasma hyopneumoniae P97抗原の一部を菌体表面に発現する豚丹毒菌YS-19株免疫豚のP97抗原に対する免疫応答の解析を行った。YS-19株を3回経鼻的に免疫された豚では,P97抗原に対する血中抗体は検出されなかったが,末梢血単核球は3回目の免疫直後からP97抗原刺激に対して増殖反応を示し,その程度は対照豚と比べて有意な差が認められた。また,M. hyopneumoniae強毒株攻撃接種後に採取した末梢血単核球はP97抗原刺激に対して細胞性免疫の重要な指標であるIFN-γを産生した。免疫,非免疫に拘わらず,豚の末梢血単核球はP97抗原の刺激によりIL-8を産生し,P97抗原を鼻腔内に噴霧投与された豚では気管支肺胞洗浄液中に多量の好中球が観察された。さらに,YS-19株のNeedle-free injector(針なし注射)を用いたワクチン接種法の検討において,皮内に1回免疫した豚の末梢血単核球は,M. hyopneumoniae強毒株攻撃接種後にP97抗原刺激による増殖能が有意に上昇した。なお,Needle-free injectorによるYS-19株免疫豚では肺炎病変の形成が有意に抑えられた。農業技術研究機構動物衛生研究所, 2007, Bulletin of the National Institute of Animal Health, (113) (113), 1 - 11, Japanese[Refereed]

- ヨーネ病の診断法に関する研究ヨーネ病診断技術の改良,開発を目的として,1)PCRによる糞便中ヨーネ菌の検出,2)インターフェロン・ガンマ(IFN-γ)検出によるヨーネ病の早期診断,3)ヨーネ菌特異抗原及び表面抗原の解析と診断への応用,4)ヨーネ菌感染における細胞性免疫応答の解析等,様々な角度からヨーネ病の新たな診断法について検討した。PCR法として,internal control DNAを添加した反応系が,サンプル毎の糞便由来PCR反応阻害作用の影響を検定することが可能であり,糞便中ヨーネ菌検出法として有用であった。IFN-γ検査では,陽性対照として設けたconcanavalin A(Con A)刺激によるIFN-γの産生量が,健康牛では最も高い値を示すのに対し,ヨーネ病感染牛ではヨーニンPPD刺激時のIFN-γ産生量が最も高い傾向が認められたことから,この点を診断上の一つの基準とすることが望ましい。ヨーネ菌の抗原解析において,類似の鳥型結核菌とは反応しないヨーネ菌に対して特異性の高いモノクローナル抗体(Mab)が得られた。さらに,当該Mabはヨーネ菌の表面に存在するリポアラビノマンナンを認識していた。ヨーネ菌Alkyl hydroperoxide reductases Cの遺伝子組換え体を作出し,本組換え抗原がヨーネ菌実験感染牛の血液細胞からIFN-γの産生を誘導することを確認した。ケモカインの一つであるmonocyte chemoattractant protein-1の産生とヨーネ菌感染との関連がマウス感染モデル実験において示唆された。農業技術研究機構動物衛生研究所, 2002, 動物衛生研究所研究報告, (109) (109), 33 - 42, Japanese

- 日本学術振興会, 科学研究費助成事業, 基盤研究(B), 東京大学, 01 Apr. 2023 - 31 Mar. 2026ルーメン細菌発電デバイスの開発本研究は、畜産動物のルーメン内環境をモニタリングするためのルーメンセンサ用の自立電源デバイスとして、ルーメン内の細菌による発電デバイスを開発することを目的としている。具体的には、ルーメン中で継続的に発電ができ、その平均発電電力が、少なくとも1時間に1回程度の無線送信と連続的なセンシングを可能とする1 mW以上となるデバイスの開発を目指す。そのために発電細菌の同定と動態解明を行うとともに、発電細菌の特性を最大限に引き出すことのできる電極構造の開発を行う。 本年度は、上記目的のため、1)発電細菌候補の絞り込み、2) 電極寿命改善のためのCNT電極の開発、3)ルーメン環境下での発電のための細菌の分離・固定化手法の検討 を行った。 1)については、長期間にわたり良好な発電を継続しているルーメン細菌叢発電の電極として使用している竹炭電極(陽極及び陰極)について、シングルセルゲノム解析を行い、細菌属のゲノム、および細胞外への電子伝達経路に関与する遺伝子群等を保有していることも確認した。 2)については、これまで良好な発電が行えていたCuペーパー電極に代えて、CNTペーパー電極を検討し、Cuペーパー電極と同等の発電量を維持しながら、電極寿命を2週間程度から半年以上に改善することに成功した。 実際のルーメン環境下で発電を持続させるためには、陽極・陰極それぞれに異なる細菌を分離・定着させる必要がある。そこで3)については、電極表面にPEDOT:PSSによって細菌を固定化する方法、および、陽極・陰極のチャンバーを分けることで細菌を分離する方法の検討を行った。

- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (A), Grant-in-Aid for Scientific Research (A), Tohoku University, Apr. 2019 - Mar. 2023内在性抗菌因子PGRPに着目した育種と飼養の抗病性シナジー強化による健全畜産戦略本年度は、以下の成果が得られた。 【課題1】子豚のPGRP2・3・4において、プロモータ及びコード領域でSNPを検出した。それらからハプロタイプの再構成を試み、PGRP2で3種、PGRP3及びPGRP4でそれぞれ7種を検出した。PGRPの遺伝型と発現の関連性を検討したところ、PGRP4の特定のハプロタイプを持つ個体について、消化器系各臓器において処置内容に関わらずPGRP4が高発現する傾向が確認された。【課題2】我々が保有するブタ由来乳酸桿菌ライブラリーより、昨年度までに評価をした菌種に加え、新たな菌種についても、ブタ小腸上皮細胞を用いてPGRPの遺伝子発現を増強するイムノバイオティクス候補菌株の評価を行った。昨年までと同様に、発現調節能は菌株特異的であり、内在性抗菌因子の強化に有用なイムノバイオティクス候補菌株の存在が再認識でき、その探索を進めることができた。【課題3】イムノバイオティクス候補菌株を7日間連続経口投与した子宮切断術作出初乳未接種(HPCD)子豚に豚ロタウイルスを経口投与した結果、ウイルス接種後3日目及び5日目の糞便中ウイルス排泄量が菌株によって対照のPBS投与群よりも有意に低かった。また、HPCD子豚末梢血中貪食細胞の化学蛍光(CL)活性は菌株投与後、FFIG67株においてはウイルス接種日あるいは接種後7日目にかけてPBS群と比較して顕著な増加が確認された。【課題4】イムノバイオティクス候補菌株を抗菌剤無添加飼料給与LWD交雑豚に投与したところ、対照区に比べ下痢の頻度が抑えられ、毒素原性大腸菌の感染は有意に低かった。さらに豚肉における総不飽和脂肪酸組成割合が有意に高くなり、枝肉格付け評価も高まった。以上のことから、イムノバイオティクス候補菌株の投与は、健康性を保ちながら産肉性の向上に寄与することが期待された。
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (B), Grant-in-Aid for Scientific Research (B), The University of Tokyo, Apr. 2019 - Mar. 2022Development of an Ultra-Long-Life Miniature Rumen pH Sensor Using a Microfluidic Control DeviceIn order to realize an ultra-long-life miniature rumen pH sensor, we realized an intermittently operating reference electrode by introducing a low power SMA valve-actuator into a liquid junction and developing a new Ag/AgCl electrode device that can redeposit AgCl. We also proposed a planar all-solid-state glass electrode structure with pressure-sensitive adhesive for ultra-long-life pH sensors, and fabricated a wireless rumen pH sensor with the prototyped glass electrode, which was fed into a fistula-forming cow for wireless communication demonstration. In addition, we investigated the electrode and mounting structure of a rumen bacterial flora power generator to introduce it into the wireless rumen pH sensor as a self-power generator.
