- Water freeze-drying has traditionally been considered an inappropriate drying method for biological samples for scanning electron microscopy (SEM) because ice crystals can potentially form and damage cell structures. Chemical fixation is also considered essential for the SEM observation of soft cells, such as protozoa and animal cells. We succeeded in preparing specimens of freshwater protists for SEM using simple water freeze-drying without chemical fixation. Specifically, a suspension of live cells in water was frozen by contact with a - 80 °C copper block using an outer cylinder that ensured precise planar contact. This significantly reduced artifact formation and increased the success rate from ~ 10% to 20-30%. Transmission electron microscopy (TEM) observations confirmed that intracellular ice crystal formation was significantly suppressed in approximately 5% of Paramecium cells, representing a notable improvement over conventional methods. This method also shortens preparation time to 2-3 h and enables high-resolution SEM imaging of various organisms. Overall, based on these findings, we standardized a simple and low-artifact method using water freeze-drying as a novel and improved technique for SEM sample preparation.Dec. 2025, Scientific reports, English, International magazineScientific journal

- Water freeze-drying of biological samples has long been considered unfeasible as a sample preparation method for scanning electron microscopy (SEM) owing to the formation of damaging ice crystals. Contrary to this assumption, however, when live ciliates in water were frozen in contact with pre-cooled copper blocks in this study, they could be freeze-dried without artifact formation, although the success rate was only about 10%. This method offers several advantages over the traditional approach of chemical fixation followed by dehydration and drying. First, the degree of sample shrinkage associated with sample preparation was much lower than with chemical fixation. Second, contractile ciliates such as Spirostomum and Lacrymaria could be sampled in their elongated state, and the metachronal waves of cilia on the cell surface were well preserved. Moreover, this method requires no special equipment and only a few hours at most for sample preparation before SEM observation. Thus, although this method needs to be improved to increase the success rate in the future, it can be used to prepare samples for SEM observation that could not be prepared by other methods.Frontiers Media SA, Nov. 2023, Frontiers in Protistology, 1Scientific journal

- We aimed to freeze-dry the ciliate Spirostomum ambiguum, obtained from water, without fixation and observe it using scanning electron microscopy (SEM). Living cells were placed on a specimen stub and frozen upon contact with a Cu block kept at either -80 °C or -100 °C. Samples were then freeze-dried and observed by SEM. In most cases, no damage to the specimen due to ice crystal formation was observed. Because of the instantaneous freezing, the metachronal wave of cilia on the body surface of the ciliate was well preserved. Approximately 70-80% of cells were preserved in the contracted state due to inevitable exposure to mechanical vibration immediately before freezing. The remaining samples were preserved in a fully-extended state. Morphometric measurements of the cell surface showed that in the extended state, ciliary rows were almost parallel to the long axis of the cell, whereas in the contracted state, they were twisted in a left-handed helix at an angle of 45-65°. The distance between adjacent cilia along a ciliary row was 1.88 ± 0.43 μm in the extended state and 1.32 ± 0.41 μm in the contracted state (mean ± SD). Abstract The genusStentor is a relatively well-known ciliate owing to its lucid trumpet shape.Stentor pyriformis represents a green, short, and fatStentor , but it is a little-known species. We investigated 124 ponds and wetlands in Japan and confirmed the presence ofS. pyriformis at 23 locations. All these ponds were noticeably oligotrophic. With the improvement of oligotrophic culture conditions, we succeeded in long-term cultivation of three strains ofS. pyriformis . The cytoplasm ofS. piriformis contains a large number of 1–3 μm refractive granules that turn brown by Lugol’s staining. The granules also show a typical Maltese-cross pattern by polarization microscopy, strongly suggesting that the granules are made of amylopectin-rich starch. By analyzing the algal rDNA, it was found that allS. pyriformis symbionts investigated in this study wereChlorella variabilis. This species is known as the symbiont ofParamecium bursaria and is physiologically specialized for endosymbiosis. Genetic discrepancies betweenC. variabilis ofS. pyriformis andP. bursaria may indicate that algal sharing was an old incident. - Freshwater protists often harbor unicellular green algae within their cells. In ciliates, possibly because of large host cell sizes and the small size of algal coccoids, a single host cell typically contains more than a hundred algal cells. While surveying such algae-bearing protists on Minami Daito Jima Island in Japan, we found a green Loxodes ciliate (Loxodida, Karyorelictea) that contained one or two dozens of very large coccoid algae. We isolated one of these algae and analyzed its characteristics in detail. A small subunit (SSU) rDNA phylogeny indicated Pseudodidymocystis species (Scenedesmaceae, Chlorophyceae) to be the taxon closest to the alga, although it was clearly separated from this by 39 or more different sites (inclusive of gaps). SSU rRNA structure analyses indicated that these displacements included eight compensatory base changes (CBCs) and seven hemi-CBCs. We therefore concluded that this alga belongs to a separate genus, and described it as Pediludiella daitoensis gen. et sp. nov. The shape of the isolated and cultured P. daitoensis was nearly spherical and reached up to 30 µm in diameter. Chloroplasts were arranged peripherally and often split and elongated. - Extant eukaryote ecology is primarily sustained by oxygenic photosynthesis, in which chlorophylls play essential roles. The exceptional photosensitivity of chlorophylls allows them to harvest solar energy for photosynthesis, but on the other hand, they also generate cytotoxic reactive oxygen species. A risk of such phototoxicity of the chlorophyll must become particularly prominent upon dynamic cellular interactions that potentially disrupt the mechanisms that are designed to quench photoexcited chlorophylls in the phototrophic cells. Extensive examination of a wide variety of phagotrophic, parasitic, and phototrophic microeukaryotes demonstrates that a catabolic process that converts chlorophylls into nonphotosensitive 132,173-cyclopheophorbide enols (CPEs) is phylogenetically ubiquitous among extant eukaryotes. The accumulation of CPEs is identified in phagotrophic algivores belonging to virtually all major eukaryotic assemblages with the exception of Archaeplastida, in which no algivorous species have been reported. In addition, accumulation of CPEs is revealed to be common among phototrophic microeukaryotes (i.e., microalgae) along with dismantling of their secondary chloroplasts. Thus, we infer that CPE-accumulating chlorophyll catabolism (CACC) primarily evolved among algivorous microeukaryotes to detoxify chlorophylls in an early stage of their evolution. Subsequently, it also underpinned photosynthetic endosymbiosis by securing close interactions with photosynthetic machinery containing abundant chlorophylls, which led to the acquisition of secondary chloroplasts. - Regulation of intracellular pH is critically important for many cellular functions. The quantification of proton extrusion in different types of cells and physiological conditions is pivotal to fully elucidate the mechanisms of pH homeostasis. Here we show the use of gold nanoparticles (AuNP) to create a high spatial resolution sensor for measuring extracellular pH in proximity of the cell membrane. We test the sensor on HepG2 liver cancer cells and MKN28 gastric cancer cells before and after inhibition of Na+/H+ exchanger. The gold surface conjugation strategy is conceived with a twofold purpose: i) to anchor the AuNP to the membrane proteins and ii) to quantify the local pH from AuNP using surface enhanced Raman spectroscopy (SERS). The nanometer size of the cell membrane anchored sensor and the use of SERS enable us to visualize highly localized variation of pH induced by H+ extrusion, which is particularly upregulated in cancer cells.Dec. 2018, Nature Communications, 9(1) (1), 5278 - 5278, English, International magazine[Refereed]Scientific journal
- Chlorella variabilis, a symbiotic alga, is usually present in the cytoplasm of Paramecium bursaria, although it can be cultured in host-free conditions. Morphological and chemical properties of its cell wall were compared between its free-living and symbiotic states. Transmission electron microscopy (quick-freezing and freeze-substitution methods) revealed that the cell wall thickness of symbiotic C. variabilis was reduced to about half that of the free-living one. Chemical properties of the cell wall were examined by treatment with three fluorescent reagents (calcofluor white M2R, FITC-WGA, and FITC-LFA) having specific binding affinities to different polysaccharides. When the algae were re-introduced into Paramecium host cells, calcofluor fluorescence intensity reduced by about 50%. Calcofluor stains β-d-glucopyranose polysaccharides such as cellulose, N-acetylglucosamine, sialic acid, and glycosaminoglycans. Because treatment with cellulase showed no effect on calcofluor fluorescence intensity, we consider that cellulose is not majorly responsible for the stainability of calcofluor. Staining intensities of FITC-WGA and FITC-LFA were similar in the free-living and symbiotic conditions, suggesting that N-acetylglucosamine and sialic acid are also not responsible for the reduction in the stainability of calcofluor associated with intracellular symbiosis. - Ants are known to use a colony-specific blend of cuticular hydrocarbons (CHCs) as a pheromone to discriminate between nestmates and non-nestmates and the CHCs were sensed in the basiconic type of antennal sensilla (S. basiconica). To investigate the functional design of this type of antennal sensilla, we observed the ultra-structures at 2D and 3D in the Japanese carpenter ant, Camponotus japonicus, using a serial block-face scanning electron microscope (SBF-SEM), and conventional and high-voltage transmission electron microscopes. Based on the serial images of 352 cross sections of SBF-SEM, we reconstructed a 3D model of the sensillum revealing that each S. basiconica houses > 100 unbranched dendritic processes, which extend from the same number of olfactory receptor neurons (ORNs). The dendritic processes had characteristic beaded-structures and formed a twisted bundle within the sensillum. At the "beads," the cell membranes of the processes were closely adjacent in the interdigitated profiles, suggesting functional interactions via gap junctions (GJs). Immunohistochemistry with anti-innexin (invertebrate GJ protein) antisera revealed positive labeling in the antennae of C. japonicus. Innexin 3, one of the five antennal innexin subtypes, was detected as a dotted signal within the S. basiconica as a sensory organ for nestmate recognition. These morphological results suggest that ORNs form an electrical network via GJs between dendritic processes. We were unable to functionally certify the electric connections in an olfactory sensory unit comprising such multiple ORNs; however, with the aid of simulation of a mathematical model, we examined the putative function of this novel chemosensory information network, which possibly contributes to the distinct discrimination of colony-specific blends of CHCs or other odor detection.2018, Frontiers in cellular neuroscience, 12, 310 - 310, English, International magazine[Refereed]Scientific journal

Paramecium bursaria is a species of ciliated protozoa which bears zoochlorellae (endosymbiotic unicellular green algae). Zoochlorellae can be artificially removed from the host cell (bleaching). Isolated zoochlorellae are able to be re-introduced to aposymbiotic P. bursaria cells (re-symbiosis). Because bleaching and re-symbiosis can easily be performed, P. bursaria is a useful model organism for the study of endosymbiosis. P. bursaria can be infected not only with zoochlorellae from the original host but also with those from different P. bursaria strains or those from different host species. Furthermore, free-living algae, yeasts and bacteria are occasionally accepted by P. bursaria. As a common characteristics of zoochlorellae, they release maltose under low-pH conditions. Zoochlorella is uniformly and closely enveloped by the peri-algal vacuole (PV) membrane, which is supposed to be indispensable for sustaining the acidic condition of the peri-algal space. PV membranes are frequently observed to be closely associated and sometimes fused with the outer membrane of host's mitochondria, suggesting that host's mitochondria are also playing an important role in the endosymbiotic event. Differential transcriptomic analysis between symbiotic and aposymbiotic cells of P. bursaria has been carried out, and some up- or down-regulated genes were identified.

- Axopodial degradation in the heliozoon Raphidiophrys contractilis: A novel bioassay system for detecting heavy metal toxicity in the aquatic environment.We observed the physiological effects of zinc, lead, mercury, copper, cadmium, and arsenic on the axopodia of the centrohelid heliozoon Raphidiophrys contractilis. In the presence of these heavy metal ions, the axopodial length of the heliozoon decreased in a concentration-dependent manner. When the heavy metal ions were examined at the same concentration, mercury produced the strongest effect on axopodia. At a high concentration (> 10-3 M) of any of the heavy metal ions examined, axopodia disappeared and cells became disrupted. Axopodia were also degraded by the addition of solutions with an acidic (< or = 6) or basic (> or = 8) pH. These observations indicate the toxic effects of heavy metal ions and non-neutral pHs on axopodial length, and also signify that R. contractilis can be used as an effective biological tool for the study of metal poisoning in eukaryotic cells.2006, Environ. - Sources of activator calcium ions in the contraction of smooth muscles in Aplysia kurodai.The physiological and pharmacological properties of contraction and the ultrastructure of buccal mass retractor muscle (I4) and gill-pinnule closure muscle (GPCM) in Aplysia kurodai were studied to learn more about the sources of activator Ca2+ in molluscan smooth muscle. Acetylcholine (ACh) and high K+-induced contractions were reduced by lowering the external Ca2+ concentration, and eliminated by the removal of extracellular Ca2+. Nifedipine appreciably reduced ACh- and high K+-induced contractions, while amiloride decreased only ACh-induced contractions and had no significant effect on high K+-induced contractions. When nifedipine and amiloride were applied together, either type of contraction was still appreciable. Serotonin (5-HT) could potentiate subsequent ACh- and high K+-induced contractions in I4; potentiated tension was significantly reduced by nifedipine and amiloride, whereas 5-HT inhibited ACh-and high K+-induced contractions in GPCM. The potentiating effects of 5-HT may be mediated by the activation of the Ca2+-channel to increase the influx from extracellular Ca2+. Caffeine caused contractions in Ca2+-free solution in both muscles. Electron microscopy revealed sarcolemmal vesicles underneath the plasma membrane in both muscle fibers. Electron microscopical cytochemistry demonstrated that pyroantimonate precipitates were localized in the sarcolemmal vesicles and in the inner surface of plasma membranes in the resting fibers. - RDB-1, a nucleolar RNA-binding protein, is essential for Caenorhadbitis elegans early development through 18S ribosomal RNA processing.RBD-1 is the Caenorhabditis elegans homolog of Mrd1p, which was recently shown to be required for 18S ribosomal RNA (rRNA) processing in yeast. To gain insights into the relationship between ribosome biogenesis and the development of multicellular organisms, we examined the expression and function of RBD-1. Maternal RBD-1 in the fertilized egg disappears immediately after cleavage starts, whereas zygotic RBD-1 first appears in late embryos and is localized in the nucleolus in most cells, although zygotic transcription of pre-rRNA is known to be initiated as early as the one-cell stage. RNA interference of the rbd-1 gene severely inhibits the processing of 18S rRNA in association with various developmental abnormalities, indicating its essential role in pre-rRNA processing and development in C.elegans. - Size distributions of intracellular organelles in an ultra-thin section are usually distorted when the section thickness is not negligible. A computer program is developed here to estimate true size distributions of spherical organelles from transmission electron micrographs of ultra-thin sections. - Ionic mechanisms were examined with reference to modification of swimming velocity and cell elongation in Triton-extracted cell models of Blepharisma. The extracted cells swam forward at Ca(2+) concentrations below 10(-6) M. The forward swimming velocity of the cell models increased with a decreased Ca(2+) concentration in the surrounding medium. At Ca(2+) concentrations above 10(-6) M, the models swam backward or rotated. The elongation of the models occurred at Ca(2+) concentrations below 10(-7) M. - An electrode sensitive to tetraphenyl phosphonium (TPP+) was applied to monitor the changes in the membrane potential of the ciliate Blepharismajaponicum during the process of cell elongation. Uptake of TPP+ by Blepharisma cells was observed when they were incubated in a mediumcontaining 2 x 10-6M TPP+. Subsequent light stimulation (3, 000 lux) led to an efflux of accumulated TPP+ from the cells, indicating that depolarization of the plasma membraneoccurred with the concomitant cell elongation. TPP+ efflux was always observed whenever cell elongation was induced in response to various stimuli: valinomycin (10-6M), K+ (30 mM), Co2+ (5 mM), and Ca2+ (5 mM). Furthermore, elongation was also evoked by electrical stimulation. - Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Kobe University, 01 Apr. 2020 - 31 Mar. 2023Search for factors enabling intracellular symbiosis using a chlorella mutant that have lost symbiotic ability原生生物ミドリゾウリムシParamecium bursariaの細胞内には数百のクロレラChlorella variabilisが共生している。このような共生関係は、二次共生の初期段階であると考えられ、細胞内共生機構を解明するためのモデルとして適している。これまでに、ミドリゾウリムシの側から共生の分子機構を調べた研究が多く報告されているが、今のところミドリゾウリムシ-クロレラの細胞内共生が成立し、さらにそれが維持される分子機構については全く分かっていない。そこで本研究では、研究の視点を変えて、クロレラ側からのアプローチによって、細胞内共生の分子機構を解明しようと試みている。本研究ではこれまでに、クロレラの細胞内共生に重要と思われるタンパク質p120についての生化学的・分子生物学的解析を行い、その遺伝子を同定することに成功した。p120はクロレラ細胞表層に存在し、宿主由来のPV膜(共生胞膜)と相互作用してクロレラのミドリゾウリムシへの共生に関連している可能性が示唆された。これまでの研究でp120はvon Willebrand Factor(vWF)と高い相同性を持つことがわかったので、ミドリゾウリムシのPV膜に存在することが想定されるp120に対するリガンドの候補を探索した。このために、ミドリゾウリムシからPV膜を単離し、質量分析によってそこに存在するタンパク質の網羅的解析を実施した。質量分析のレファレンスデータとしては、過去の研究によって取得したミドリゾウリムシのmRNA-Seqデータを利用した。

- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Nara Women's University, 01 Apr. 2013 - 31 Mar. 2017The function of mating pheromone and the speciation in BlepharismaTo investigate the function and the action mechanism of sex pheromone (gamone) in ciliate Blepharisma, ①Isolated gamone 1 genes from several stocks of Blepharisma and identified the sites which determine the specificity of species, ② examined the localization of gamone receptors in the cells, ③ tried to identify genes involved in biosynthesis of gamone 2. In order to clarify the phylogenetic relationship of the strains of Blepharisma, ④ isolated several genes and constructed phylogenetic trees, and ⑤ investigated the specificity of gamone 1 between species, whether the pair formation between different megakaryotype strains and between different species occrred, and whether progenies between such crosses survived, and reexamined the classification of the genus Blepharisma.

At first, kendrin and Cep152 can interact via their carboxyl terminals. Second, although Cep152 may not localize at the centrosome during G2/M phase they were recruited to the centrosome again during cytokinesis. Third, by siRNA mediated- kendrin depletion, expression level of Cep152 were increased and Cep152 could localize at the centrosome efficiently rather than normal condition. Taken together I found that Cep152 might be recruited efficiently to the centrosome after kendrin was released and that this mechanism could facilitate centrosome duplication at next cell cycle.

We executed continuous sampling of the ciliates and their algal DNA analyses. In consequence, we found that comparatively large number of ciliate species maintain their own algae, and the small number of algal species are shared among different host species. It was found some differences, e.g., nucleotide substitutions in spacer region, in the same species of algae among different host species, which may also indicate the persistent symbioses.

Plastids and mitochondria are derived from endosymbiosis between bacteria and ancestral eukaryotes (most likely archeae), and gave significant metabolic advantages to eukaryotes. This study aimed at (1) identification of various primary and secondary (endo)symbiosis in nature and elucidation of shared fundamental mechanisms, (2) elucidation of physiological functions and mechanisms of quality control of such endosymbiosis-derived organelles, (3) establishment of a new paradigm of “endosymbiosis-driven evolution of eukaryotes”, which we named “Matryoshka-type evolution”, and (4) development of synthetic biological methods that allow artificial evolution of cell functions, such as addition of new metabolic functions to organelles and organelle manipulation/transfer.

The results indicate that R128 is one of the key residues preserving the ability to recognize all three stop codons, especially UGA in ciliate eRF1s.

Therefore, we, more than 20 protozoan specialists, have conducted a systematic survey on the Japanese protozoans by 1) collecting original bibliographic records 2) listing up species names and collection points, and 3) extracting reliable list of literature, in order to construct a comprehensive inventory database of Protozoan fauna of Japan. We have also conducted a nation -wide field survey of free-living and parasitic protozoans at more than 100 locations in Japan, including Abashiri area in Hokkaido and Minami Daito Island in Okinawa prefecture. - Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (B), Hiroshima University, 2008 - 2012Studies on life cycles of aquatic parasites, in particular, host-specificities of their infective stagesThe life cycle, growth and reproduction of Caligus fugu(=Pseudocaligus fugu), which causes “caligosis” in tiger puffer, are clarified. In caligiforms the following results were newly obtained: (1) the number of post-naupliar stages is six as in other copepods; (2) some species have a life cycle to switch hosts in adults; (3) the suck-like lunule developmentally originates from the cuticle membrane of frontal plates. The monogenean Udonella fugu hyperparasitic on C. fugu on glass puffer occurred and reproduced throughout a year. Three species of trematodes utilize large-sized medusae, medusivorous fish and Cyanea nozakii as the second, final and paratenic hosts, respectively. The histophagous apostome ciliate Vampyrophrya pelagicametamorphoses from tomite to phront, during which cyst wall and stalk and precursors of food vacuole membranes are formed.

- 科学技術振興機構, 産学が連携した研究開発成果の展開/研究成果展開事業/研究成果最適展開支援プログラム(A-STEP)/探索タイプ, 2010 - 2010超小型水質バイオモニタリング装置に用いるためのハリタイヨウチュウの大量培養技術とカートリッジへの注入・保持・移送技術の開発原生動物ハリタイヨウチュウを用いた水質モニタリング装置に用いるための、ハリタイヨウチュウの大量培養方法と、カートリッジへ装填する手法を検討した。加えて、ユーザーの手元にまで輸送するための方法を検討した。研究の具体的な内容としては、1)神戸大学において、ハリタイヨウチュウの品質安定大量培養系を確立した。そこ結果、培養液の組成・温度・餌の種類を検討することで、これまでの約10倍効率的で、品質の安定した培養法を確立した。2)岡山大学では、脱気装置・接続プラグ等を改良し、ハリタイヨウチュウの測定カートリッジへの充填法などを改良し、安全に運搬するための輸送容器の設計を行った。
- Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Grant-in-Aid for Scientific Research (C), Nara Women's University, 2006 - 2008Study on the expression and function of sex pheromone in ciliates原生生物繊毛虫Blepharismaの接合誘導時特異的に発現する遺伝子を単離・同定した。糖鎖を欠く接合誘導物質ガモン1を(1) 酵素処理、および(2) 大腸菌を用いた大量発現系により得て、ガモン1の糖鎖は誘導活性に重要な役割をもつことを明らかにした。ガモン1に対する抗体を作製し、II型細胞表面にあるガモン1受容体の探索を行った。野外で採集したBlepharismaの種を同定し、rRNA遺伝子とガモン1遺伝子の配列を調べ、Blepharismaの種の分岐について考察した。

Completion of its life cycle usually needs feeding by invertebrate predators such as chaetognaths on copepods with phoronts of the ciliates. In contrast predation by fish larvae was not involved in it. Cilia of the ciliates are formed within the phoront, which guarantees quick excystation. Numerous thick membranes detected in the cytoplasm are considered to be precursors of food vacuole membranes of the trophont, and totally disappeared in the fully-grown cell. The morphology of the cytostome was newly revealed with SEM and TEM. Relationships between water temperature and duration of each stage of the ciliate were investigated in the laboratory. The maximum time from phoront hatching to tomite appearance at the lowest temperature (10℃) was nearly 20 times as long as the minimum time at the highest temperature (25℃), which seems to be caused by prolongation of the tomont. Apostome phoronts were broadly found from copepods collected from oceanic islands, deep-sea hyperbenthic layers, and brackish waters. This strongly suggests their paramount importance in marine food webs. A nicothoid copepod and an epicaridean isopod were newly described from the marsupium of the mysid Siriella okadai in the Seto Inland Sea. The maximum incidences of these parasites were up to 11 and 19 %, respectively. These parasites may compete severely with the same habitat and food. Intersex individuals of the shrimp Acetes sibogae and the mysid Siriella japonica izuensis were discovered, in the latter of which it seems to have been caused by infection of ellobiopsids. The occurrence of the dinoflagellate Oodinium inlandicum on pelagic chaetognaths in Korean and of the ellobiopsid Thalassomyces marsupii on the amphipod Scina vosseleri were also reported for the first time.

In consequence, this research project has been carried out for two years starting from the academic year of 1994. In the first academic year, five sequential programs were successfully produced by the aid of N88BASIC language using video disc recorder of postscript type. In this case, that is to say, program #1 was for a content showing the other programs. #2 for a true form of retrieval program, #3 for a program of retrieval tables on the basis of two types of files for retrieval data and for protozoan taxonomy, #4 for producing a flow chart of retrieval based on the data file, and finally #5 for transforming the files into random access type by making rapid retrieval possible. In the second academic year (1995), this research project was focused on producing video image library of a number of species of Protozoa using differential interference optics of a light microscope. As the result, a video software for retrieving protozoan species became possible to utilize, although it is written in Japanese at the present time.
- 日本学術振興会, 科学研究費助成事業, 試験研究, 高知医科大学, 1987 - 1988生体臓器・組織・細胞を対象とする広帯域誘電解析法の開発と標準化〈測定セルの開発と試作〉 微小量の細胞サスペンションを測定するためのセルを試作した。浮遊容量・残留インダクタンスともに、従来型のセルに比べて約1/10であり、アーチファクト補正時の誤差をかなり小さくすることができた。また、インピーダンス・アナライザとの適合性に極力留意したので、スプリングクリップフィクスチュアを介して瞬時に脱着可能となった。 高周波域の測定を1GHzまで拡張する目的で、インダクタンス補正を要しない同軸型セルを開発した。これにより、現有のRF4191Aアナライザの性能をフルに活用できるようになった。これはサスペンション系の高周波域での測定にとり今後威力を発揮するものと期待される新しいセルである。さらに、眼レンズおよび筋組織測定用のセルも試作した。 〈測定・解析のためのコンピュータプログラミング〉 前年度において開発したプログラムをより使い易くかつ高速化できるように種々の改良をおこなった。これにより4194A型アナライザを用いるばあい10KHz〜100MHz域での81点の測定が25秒以内に完了することとなった。これは生細胞を扱う際大きな利点となるものである。 解析プログラムでは、最も単純なOne-Shellモデルにはじまり、一般の細胞を念頭においたVesicle-Inclusion Three-Shellモデルに至る合計9種類のモデル解析用の解析システムが完成した。これは京大化学研究所、広島大総合科学部等の研究者にも分与され、大いに活用されている。 〈生体試料の誘電解析〉 細胞系では、赤血球、培養白血病細胞などの厳密なモデル解析をおこない、従来通説となっていた界面分極理論に対して新しい問題提起をおこなった。水晶体および横紋筋の解析では組織インピーダンに関する新解釈を提出すべく目下研究を続行中である。
- 日本学術振興会, 科学研究費助成事業, 一般研究(C), 高知医科大学, 1987 - 1988小腸上皮細胞における糖・アミノ酸輸送能とPHiおよびCaiとの関連小腸繊毛組織における上皮細胞のNaと糖やアミノ酸との共輸送活性には、上皮細胞内pHおよびCa濃度の意動に応じてそれぞれの活性調節がなされているものと想像される。in situの細胞動態を追究する前段階の作業として、ニワトリ小腸粘膜より単離した上皮細胞(エンテロサイト)の懸濁系を材料として主に蛍光プローブ法により、pHiの調節に関係する種々のイオン輸送系の基本的性質を調べた。 pHiの測定はアクリジンオレンジの蛍光変化を指標して実施した。 120mMNa^+を主体とする30℃の生理的メジウム条件の下で、エンテロサイト膜にNa^+/H^+交換輸送系が存在しており、有意に機能している思われる知見が得られた。 重炭酸イオンを含まない条件下においても、細胞外液のアニオンを全て硫酸イオンに置きかえた場合、細胞内塩素イオンに由来するcl^-の外向き勾配に駆動されるpHiの上昇が観察された。種々の阻害剤を用いた実験から、このpHi上昇は、Cl^+/OH^-(またはHCO_3^-)交換輸送系のはたらきによるものを判定した。 同じく阻害剤の実験から、上に述べたNa^+/H^+およびCl^-/OH┣D1-(HCO┣D23┫D2┣D1-┫D1)交換輸送系の作動のみでは説明できないpHi上昇などの変化を見出した。これは腎尿細管細胞の膜において近年見出されてきているNa┣D1+┫D1依存性Hco┣D23┫D2┣D1-┫D1/Cl┣D1-┫D1交換輸送系と同様のものであろうと考えられる。 以上の如く、少なくとも3種のpHi調節に関与する輸送系の存在することをニワトリの単離エンテロサイトにおいて確認した。
